Modelling ⁸⁵Kr datasets with python for applications in tracer hydrology and to investigate atmospheric circulation.

ABSTRACT: We present a model written in python to evaluate data from comprehensive ⁸⁵Kr collection schemes comprising 11 datasets from different monitoring stations around the globe. The model is designed to (1) calculate atmospheric input functions for the application of ⁸⁵Kr as a dating tracer and (2) to investigate atmospheric circulation based on a two-box model of the atmosphere. Different functions were implemented, to (1) filter the data, (2) fit polynomials and running means, (3) extrapolate fits from the northern to the southern hemisphere, (4) calculate interhemispheric exchange times and ⁸⁵Kr emission rates and (5) export data to a csv file. Although the model is designed to evaluate atmospheric ⁸⁵Kr datasets, some functionality and basic concepts can be applied to other dating tracers, like tritium and SF₆.•Standardized method to systematically analyse atmospheric ⁸⁵Kr activity concentration time series for dating water and ice and to investigate atmospheric circulation.•Easily modifiable python script to adapt functions for similar data analysis procedures.

Project description:The interplay between microbes and atmospheric physical and chemical conditions is an open field of research that can only be fully addressed using multidisciplinary approaches. The lack of coordinated efforts to gather data at representative temporal and spatial scales limits aerobiology to help understand large scale patterns of global microbial biodiversity and its causal relationships with the environmental context. This paper presents the sampling strategy and analytical protocols developed in order to integrate different fields of research such as microbiology, -omics biology, atmospheric chemistry, physics and meteorology to characterize atmospheric microbial life. These include control of chemical and microbial contaminations from sampling to analysis and identification of experimental procedures for characterizing airborne microbial biodiversity and its functioning from the atmospheric samples collected at remote sites from low cell density environments. We used high-volume sampling strategy to address both chemical and microbial composition of the atmosphere, because it can help overcome low aerosol and microbial cell concentrations. To account for contaminations, exposed and unexposed control filters were processed along with the samples. We present a method that allows for the extraction of chemical and biological data from the same quartz filters. We tested different sampling times, extraction kits and methods to optimize DNA yield from filters. Based on our results, we recommend supplementary sterilization steps to reduce filter contamination induced by handling and transport. These include manipulation under laminar flow hoods and UV sterilization. In terms of DNA extraction, we recommend a vortex step and a heating step to reduce binding to the quartz fibers of the filters. These steps have led to a 10-fold increase in DNA yield, allowing for downstream omics analysis of air samples. Based on our results, our method can be integrated into pre-existing long-term monitoring field protocols for the atmosphere both in terms of atmospheric chemistry and biology. We recommend using standardized air volumes and to develop standard operating protocols for field users to better control the operational quality.

Project description:Atmospheric circulation patterns derived from multi-spectral remote sensing can serve as a guide for choosing a suitable entry location for a future in situ probe mission to the Ice Giants. Since the Voyager-2 flybys in the 1980s, three decades of observations from ground- and space-based observatories have generated a picture of Ice Giant circulation that is complex, perplexing, and altogether unlike that seen on the Gas Giants. This review seeks to reconcile the various competing circulation patterns from an observational perspective, accounting for spatially-resolved measurements of: zonal albedo contrasts and banded appearances; cloud-tracked zonal winds; temperature and para-H2 measurements above the condensate clouds; and equator-to-pole contrasts in condensable volatiles (methane, ammonia, and hydrogen sulphide) in the deeper troposphere. These observations identify three distinct latitude domains: an equatorial domain of deep upwelling and upper-tropospheric subsidence, potentially bounded by peaks in the retrograde zonal jet and analogous to Jovian cyclonic belts; a mid-latitude transitional domain of upper-tropospheric upwelling, vigorous cloud activity, analogous to Jovian anticyclonic zones; and a polar domain of strong subsidence, volatile depletion, and small-scale (and potentially seasonally-variable) convective activity. Taken together, the multi-wavelength observations suggest a tiered structure of stacked circulation cells (at least two in the troposphere and one in the stratosphere), potentially separated in the vertical by (i) strong molecular weight gradients associated with cloud condensation, and by (ii) transitions from a thermally-direct circulation regime at depth to a wave- and radiative-driven circulation regime at high altitude. The inferred circulation can be tested in the coming decade by 3D numerical simulations of the atmosphere, and by observations from future world-class facilities. The carrier spacecraft for any probe entry mission must ultimately carry a suite of remote-sensing instruments capable of fully constraining the atmospheric motions at the probe descent location.
