Stem cell-derived mouse embryos develop within an extra-embryonic yolk sac to form anterior brain regions and a beating heart

Project description:We report the analysis by inDrop single cell sequencing of mouse embryos at embryonic day E6.5, E7.5 and E8.5 as well as the analysis of stem cell derived synthetic embryos generated with the previously reported protocol (Amadei et al., 2021) and cultured to day 5, day 6 and day 8 of development. We find that all the cell types present in the natural embryos examined at these timepoints were also present in their synthetic counterparts. Furthermore, in these synthetic structures we observed formation of neuroectoderm and surface ectoderm, a beating heart, gut and primordial germ cells and development occurred inside extraembryonic membranes (yolk sac and amnion) similarly to what occurs in natural development. We conclude that these synthetic embryos are a unique tool to recapitulate mouse post-implantation development.

Investigations on the role of Blimp1 in pluripotency and reprogramming

Project description:Blimp1 (Prdm1), the key determinant of primordial germ cells (PGC), plays a combinatorial role with Prdm14 during PGC specification from postimplantation epiblast cells. They together initiate epigenetic reprogramming in early germ cells towards an underlying pluripotent state, which is equivalent to embryonic stem cells (ESC). Whereas Prdm14 alone can promote reprogramming and is important for the propagation of the pluripotent state, it is not known if Blimp1 is similarly involved. Using a genetic approach, we demonstrate that Blimp1 is dispensable for the derivation and maintenance of ESC and postimplantation epiblast stem cells (EpiSC). Notably, Blimp1 is also dispensable for reprogramming EpiSC to ESC. Thus, while Blimp1 is obligatory for PGC specification, it is not required for the reversion of EpiSC to ESC, and for their maintenance thereafter. This study suggests that reprogramming, including that of somatic cells to ESC, may not entail an obligatory route through Blimp1 positive PGC-like state.
