Project description:Milk production during the early postpartum in dairy cows can impact the circulating metabolites and negatively affects embryo survival. However, the molecular consequences for the embryo during its development in the oviduct are still unknown. The objective was to determine the impact of metabolic status on embryonic genome activation (EGA) using high-throughput sequencing to generate comprehensive transcriptome profiles of bovine 16-cell stage embryos which had undergone EGA in vitro or in the oviducts of primiparous postpartum dry or postpartum lactating Holstein Friesian cows.

Proteomic analysis of the human milieu of gamete /embryo maternal interaction

Project description:The improvement of the embryo culture media is of high relevance due to its influence on successful implantation rates, pregnancy, neonatal outcomes and potential effects in the adult life. The ideal conditions for embryo development are those naturally occurring in the female reproductive tract, i.e., the oviductal and uterine fluids. In order to shed light on the differences between chemical and natural media, we performed the first comparative study of the low abundance proteins in plasma, uterine and oviductal fluid collected, simultaneously, from healthy and fertile women that underwent a salpingectomy. The rationale for this design derives from the fact that high-abundant proteins in these fluids are usually those coming from blood serum and frequently mask the detection of low abundant proteins with a potential significant role on specific processes related to the embryo-maternal interaction. The proteomic analysis by 1D-nano LC ESI-MSMS detected several proteins in higher amounts in oviductal fluid when compared to uterine and plasma samples (RL3, GSTA1, EZRI, DPYSL3, GARS, HSP90A). Such oviductal fluid proteins could be a target to improve fertilization rates and early embryo development, if used in the culture media. In conclusion, this study presents a high-throughput analysis of female reproductive tract fluids and contributes to the knowledge of oviductal and uterine secretome.

Paternal under and over nutrition modify semen composition and preimplantation embryo developmental kinetics in mice

Project description:Poor parental health can influence the development and eventual health outcomes of their offspring. Many studies have detailed the maternal role yet a father’s health requires further examination as his poor nutritional status has also been shown to have negative consequences on fetal development and impact the long-term health of his offspring. The present study examined how preimplantation embryo development was altered by sub-optimal paternal diet, with specific focus on sperm- and seminal plasma-mediated mechanisms. To address this, male mice were fed a diet to model either under (low protein diet (LPD)) or over (high fat/sugar ‘Western’ diet (WD)) nutrition, LPD or WD supplemented with methyl-donors or a control diet (CD) before mating. Embryo development was assessed using in vitro time-lapse imaging of preimplantation embryos which revealed a significant increase in embryo development rates in all experimental groups when compared to CD embryos. Further analysis of the semen revealed a significant number of differentially expressed seminal plasma proteins in all groups (LPD: 13, MD-LPD: 27, WD: 24, MD-WD: 19) when compared to control. This study highlights a role for paternal nutritional status influencing embryonic development and the maternal reproductive tract via changes to his metabolic and reproductive health. These findings demonstrate potential direct (sperm-mediated) and indirect (seminal plasma-mediated) pathways in which a father's poor diet could shape the long-term health of his offspring

Project description:During the initial maternal recognition of pregnancy (MRP), the equine embryo displays a series of unique events characterized by rapid blastocyst expansion, secretion of a diverse array of molecules, and transuterine migration to interact with the luminal epithelium surface. Up to date, the intricate transcriptome and proteome changes of the embryo underlying these events have not been critically studied in horses. Thus, the objective of this study was to perform an integrative transcriptomic and proteomic analyses of embryos collected from days 10 to 13 of gestation.

Maternal Smchd1 regulates Hox gene expression and patterning in the mouse embryo [ESC RRBS]

Project description:Parents transmit genetic and epigenetic information to their offspring. Maternal effect genes regulate the offspring epigenome to ensure normal development. Here we report that the epigenetic regulator SMCHD1 has a maternal effect on Hox gene expression and skeletal patterning. Maternal SMCHD1, present in the oocyte and preimplantation embryo, prevents precocious activation of Hox genes post-implantation. Without maternal SMCHD1, highly penetrant posterior homeotic transformations occur in the embryo. Hox genes are decorated with Polycomb marks H2AK119ub and H3K27me3 from the oocyte throughout early embryonic development; however, loss of maternal SMCHD1 does not deplete these marks. Therefore, we propose maternal SMCHD1 acts downstream of Polycomb marks to establish a chromatin state necessary for persistent epigenetic silencing and appropriate Hox gene expression later in the developing embryo. This is a striking role for maternal SMCHD1 in long-lived epigenetic effects impacting offspring phenotype.

Maternal diabetes alters transcriptional programs in the developing embryo.
