HeLa S3 cells were treated with 3.3 µM nocodazole (Noc) for 1 hour. Cells were labeled with Ska antibodies (red) and CREST antibodies (green), and DNA was labeled with DAPI (blue).

HeLa S3 cells were transfected with Myc-Ska1-4A and mCherry-Ska3-3A and fixed after 10 hour release from the second thymidine block. Cells were placed at 4ºC for 20 min before being labeled with mCherry (red), tubulin antibody (green), and DAPI (blue).

HeLa S3 cells were transfected with a plasmid encoding wild-type Mis12-INCENP-GFP. Cells were labeled with Ska3 antibodies (red), and Mis12-INCENP-GFP and DAPI were visualized by GFP fluorescence (green) and DAPI (blue), respectively.

HeLa S3 cells were co-transfected with a plasmid encoding wild-type Mis12-INCENP-GFP and with the phosphomimetic Ska plasmids Myc-Ska1-4S and mCherry-mSka3-3D. mCherry-Ska3-3D, Mis12-INCENP-GFP, and DAPI were visualized with mCherry (red), GFP (green), and DAPI (blue), respectively.

HeLa S3 cells were co-transfected with a plasmid encoding wild-type Mis12-INCENP-GFP and with non-phosphorylatable Ska plasmids Myc-Ska1-4A and mCherry-Ska3-3A. mCherry-Ska3-3A, Mis12-INCENP-GFP, and DAPI were visualized with mCherry (red), GFP (green), and DAPI (blue), respectively.

HeLa S3 cells were transfected with wild-type Myc-Ska1 and mCherry-Ska3 and fixed after 10 hour release from a second thymidine block. Cells were placed at 4ºC for 20 min before being labeled with mCherry (red), tubulin antibody (green), and DAPI (blue).
