Prior to experimental manipulation, explants were equilibrated for at least 1 h at 37 °C in a humidified atmosphere containing 5% CO₂. Explants were cultured in 12-well plates for up to 5 days in a proprietary, chemically defined, hydrocortisone- and serum-free medium supplemented with penicillin (100 µg/mL) and streptomycin (100 µg/mL). Cultures were maintained at 37 °C and 5% CO₂, and culture medium was replaced daily. Skin explants were treated topically once daily with either vehicle control (Laboratoire Gola, Galenicare SA) or VTAMA (tapinarof cream, 1%, Dermavant Sciences) throughout the culture period.</sample_protocol><sample_protocol>Library Construction - Illumina TruSeq Stranded mRNA protocol (Directional, first strand)</sample_protocol><sample_protocol>Sequencing - The mRNA was sequenced by the Next Generation Sequencing Platform of Bern using paired-end 100 bp sequencing.</sample_protocol><sample_protocol>Nucleic Acid Extraction - Total RNA from Genoskin samples was isolated using the RNeasy Lipid Tissue Kit (Cat. 74804, Qiagen) according to the manufacturer’s instructions.</sample_protocol><figure_sub>Organization</figure_sub><figure_sub>MINSEQE Score</figure_sub><figure_sub>Assays and Data</figure_sub><figure_sub>MAGE-TAB Files</figure_sub><pubmed_authors>Nicole Leonie Bertschi</pubmed_authors></additional><is_claimable>false</is_claimable><name>RNA-seq of human Genoskin explants treated with topical VTAMA (tapinarof) against untreated control</name><description>Effect of topical Tapinarof (VTAMA) on human Genoskin skin explants.
