Here, we report that activation of cyclic GMP-AMP synthase (cGAS) diminishes cognitive resilience by decreasing the neuronal transcriptional network of myocyte enhancer factor 2c (MEF2C) through type I interferon (IFN-I) signaling. Pathogenic tau activates cGAS and IFN-I responses in microglia, in part mediated by cytosolic leakage of mitochondrial DNA. Genetic ablation of Cgas in mice with tauopathy diminished the microglial IFN-I response, preserved synapse integrity and plasticity and protected against cognitive impairment without affecting the pathogenic tau load. cGAS ablation increased, while activation of IFN-I decreased, th</pubmed_abstract><journal>Nature neuroscience</journal><pubmed_title>Tau activation of microglial cGAS-IFN reduces MEF2C-mediated cognitive resilience.</pubmed_title><pmcid>PMC10166855</pmcid><funding_grant_id>5K12HD052896</funding_grant_id><funding_grant_id>A20201312F</funding_grant_id><funding_grant_id>K01 AG057862</funding_grant_id><funding_grant_id>R01AG064239</funding_grant_id><funding_grant_id>R01AG072758</funding_grant_id><funding_grant_id>K12 HD052896</funding_grant_id><funding_grant_id>T32 HD098061</funding_grant_id><funding_grant_id>R01 AG074541</funding_grant_id><funding_grant_id>R01AG054214</funding_grant_id><funding_grant_id>R01 AG054214</funding_grant_id><funding_grant_id>R01AG074541</funding_grant_id><funding_grant_id>R01 AG072758</funding_grant_id><funding_grant_id>R01 AG064239</funding_grant_id><pubmed_authors>Gong S</pubmed_authors><pubmed_authors>Corona C</pubmed_authors><pubmed_authors>Raju RM</pubmed_authors><pubmed_authors>Zhao M</pubmed_authors><pubmed_authors>Yarahmady A</pubmed_authors><pubmed_authors>Sinha SC</pubmed_authors><pubmed_authors>Kauwe G</pubmed_authors><pubmed_authors>Fan L</pubmed_authors><pubmed_authors>Mok SA</pubmed_authors><pubmed_authors>Nagiri RK</pubmed_authors><pubmed_authors>Wong MY</pubmed_authors><pubmed_authors>Torres ERS</pubmed_authors><pubmed_authors>Tracy TE</pubmed_authors><pubmed_authors>Udeochu JC</pubmed_authors><pubmed_authors>Lo I</pubmed_authors><pubmed_authors>Carling GK</pubmed_authors><pubmed_authors>McGurran H</pubmed_authors><pubmed_authors>Coronas-Samano G</pubmed_authors><pubmed_authors>Mousa GA</pubmed_authors><pubmed_authors>Gill MT</pubmed_authors><pubmed_authors>Huang Y</pubmed_authors><pubmed_authors>Ye P</pubmed_authors><pubmed_authors>Ratan RR</pubmed_authors><pubmed_authors>Gan L</pubmed_authors><pubmed_authors>Amin S</pubmed_authors><pubmed_authors>Luo W</pubmed_authors><pubmed_authors>Liu B</pubmed_authors><pubmed_authors>Tsai LH</pubmed_authors><pubmed_authors>Holtzman J</pubmed_authors></additional><is_claimable>false</is_claimable><name>Tau activation of microglial cGAS-IFN reduces MEF2C-mediated cognitive resilience.</name><description>Pathological hallmarks of Alzheimer's disease (AD) precede clinical symptoms by years, indicating a period of cognitive resilience before the onset of dementia.
