However, the longitudinal epilepsy course and developmental endpoints have not yet been described in detail, which is a critical prerequisite for clinical trial readiness. Here, we assessed 1,281 cumulative patient-years of seizure and developmental histories in 162 individuals with STXBP1-related disorders and established a natural history framework. STXBP1-related disorders are characterized by a dynamic pattern of seizures in the first year of life and high variability in neurodevelopmental trajectories in early childhood. Epilepsy onset differed across seizure types, with 90% cumulative onset for infantile spasms by 6 months and focal-onset seizures by 27 months </pubmed_abstract><journal>medRxiv : the preprint server for health sciences</journal><pagination>2023.05.10.23289776</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC10197795</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Delineating clinical and developmental outcomes in STXBP1-related disorders.</pubmed_title><pmcid>PMC10197795</pmcid><funding_grant_id>R01 NS131512</funding_grant_id><funding_grant_id>K02 NS112600</funding_grant_id><funding_grant_id>R01 NS127830</funding_grant_id><funding_grant_id>U54 HD086984</funding_grant_id><funding_grant_id>UL1 TR001878</funding_grant_id><pubmed_authors>McKee J</pubmed_authors><pubmed_authors>Prosser B</pubmed_authors><pubmed_authors>Cunningham K</pubmed_authors><pubmed_authors>Thalwitzer KM</pubmed_authors><pubmed_authors>Kaufman MC</pubmed_authors><pubmed_authors>Rigby CS</pubmed_authors><pubmed_authors>Goss J</pubmed_authors><pubmed_authors>Fitch E</pubmed_authors><pubmed_authors>Pierce SR</pubmed_authors><pubmed_authors>Ruggiero SM</pubmed_authors><pubmed_authors>Sullivan KR</pubmed_authors><pubmed_authors>Helbig I</pubmed_authors><pubmed_authors>Boland M</pubmed_authors><pubmed_authors>Syrbe S</pubmed_authors><pubmed_authors>Xian J</pubmed_authors><pubmed_authors>Toib J</pubmed_authors><pubmed_authors>Fitter N</pubmed_authors><pubmed_authors>Brimble E</pubmed_authors><pubmed_authors>deCampo D</pubmed_authors></additional><is_claimable>false</is_claimable>
Delineating clinical and developmental outcomes in STXBP1-related disorders.

STXBP1-related disorders are among the most common genetic epilepsies and neurodevelopmental disorders.
