Here, building on our prior work that established the suitable non-basal medium components for hiPSC growth, we develop a simplified basal medium consisting of just 39 components, demonstrating that many ingredients of DMEM/F12 are either not essential or are at suboptimal concentrations. This new basal medium along with the supplement, which we call BMEM, enhances the growth rate of hiPSCs over DMEM/F12-based media, supports derivation of multiple hiPSC lines, and allows differentiation to multiple lineages. hiPSCs cultured in BMEM consistently have enhanced expression of undifferentiated cell markers such as POU5F1 and NANOG, along with increased expression of markers of</pubmed_abstract><journal>Stem cell reports</journal><pubmed_title>Nutritional requirements of human induced pluripotent stem cells.</pubmed_title><pmcid>PMC10277817</pmcid><funding_grant_id>R01 CA261898</funding_grant_id><funding_grant_id>R01 CA220002</funding_grant_id><pubmed_authors>Pongpamorn P</pubmed_authors><pubmed_authors>Gharib M</pubmed_authors><pubmed_authors>Lyra-Leite DM</pubmed_authors><pubmed_authors>Javed H</pubmed_authors><pubmed_authors>Sapkota Y</pubmed_authors><pubmed_authors>Shah D</pubmed_authors><pubmed_authors>Burridge PW</pubmed_authors><pubmed_authors>Weddle CJ</pubmed_authors><pubmed_authors>Copley RR</pubmed_authors><pubmed_authors>Freeman PP</pubmed_authors><pubmed_authors>Lenny B</pubmed_authors><pubmed_authors>Fonoudi H</pubmed_authors><pubmed_authors>McKenna DE</pubmed_authors><pubmed_authors>Pinheiro EA</pubmed_authors></additional><is_claimable>false</is_claimable><name>Nutritional requirements of human induced pluripotent stem cells.</name><description>The nutritional requirements for human induced pluripotent stem cell (hiPSC) growth have not been extensively studied.
