Silencing information regulator 1 (SIRT1) was demonstrated to modulate cholesterol and lipid metabolism in NAFLD. Here, a novel SIRT1 activator, E1231, was studied for its potential improvement effects on NAFLD. C57BL/6J mice were fed a high-fat and high-cholesterol diet (HFHC) for 40 weeks to create a NAFLD mouse model, and E1231 was administered by oral gavage (50 mg/kg body weight, once/day) for 4 weeks. Liver-related plasma biochemistry parameter tests, Oil Red O staining, and hematoxylin-eosin staining results showed that E1231 treatment ameliorated plasma dyslipidemia, plasma marker levels of liver damage (alanine aminotransferase (ALT) and aspartate aminotransferase (AST)), liver total cholesterol (TC</pubmed_abstract><journal>Current issues in molecular biology</journal><pubmed_title>SIRT1 Activator E1231 Alleviates Nonalcoholic Fatty Liver Disease by Regulating Lipid Metabolism.</pubmed_title><pmcid>PMC10297637</pmcid><funding_grant_id>CPA-B04-ZC-2021-005</funding_grant_id><funding_grant_id>2022-JKCS-10, 2021-1-I2M-030</funding_grant_id><funding_grant_id>81973328</funding_grant_id><pubmed_authors>Lei L</pubmed_authors><pubmed_authors>Zhang J</pubmed_authors><pubmed_authors>Si S</pubmed_authors><pubmed_authors>Li Y</pubmed_authors><pubmed_authors>Li S</pubmed_authors><pubmed_authors>Zhang Y</pubmed_authors><pubmed_authors>Jiang X</pubmed_authors><pubmed_authors>Wang W</pubmed_authors><pubmed_authors>Han J</pubmed_authors><pubmed_authors>Liu C</pubmed_authors><pubmed_authors>Sheng R</pubmed_authors><pubmed_authors>Wu Y</pubmed_authors><pubmed_authors>Xu Y</pubmed_authors></additional><is_claimable>false</is_claimable><name>SIRT1 Activator E1231 Alleviates Nonalcoholic Fatty Liver Disease by Regulating Lipid Metabolism.</name><description>Nonalcoholic fatty liver disease (NAFLD) is one of the most common liver diseases.
