Radiation therapy remains the only effective treatment yet provides a 5-year survival rate of only 1%. Several clinical trials have attempted to enhance radiation antitumor activity using radiosensitizing agents, although none have been successful. Given this, there is a critical need for identifying effective therapeutics to enhance radiation sensitivity for the treatment of DMG. Using high-throughput radiosensitivity screening, we identified bromo- and extraterminal domain (BET) protein inhibitors as potent radiosensitizers in DMG cells. Genetic and pharmacologic inhibition of BET bromodomain activity reduced DMG cell proliferation and enhanced radiation-induced DNA damage by inhibiting DNA repair </pubmed_abstract><journal>The Journal of clinical investigation</journal><pubmed_title>BET bromodomain inhibition potentiates radiosensitivity in models of H3K27-altered diffuse midline glioma.</pubmed_title><pmcid>PMC11213469</pmcid><funding_grant_id>P50 CA221747</funding_grant_id><funding_grant_id>N/A</funding_grant_id><funding_grant_id>22IC36</funding_grant_id><funding_grant_id>R01 NS126513</funding_grant_id><funding_grant_id>R01CA197313</funding_grant_id><funding_grant_id>5P50CA221747</funding_grant_id><funding_grant_id>R01NS126513</funding_grant_id><funding_grant_id>R01 CA277605</funding_grant_id><funding_grant_id>R01 CA258636</funding_grant_id><funding_grant_id>R01CA258636</funding_grant_id><funding_grant_id>R01 CA197313</funding_grant_id><funding_grant_id>R01 CA204297</funding_grant_id><funding_grant_id>R01 NS093079</funding_grant_id><funding_grant_id>863193</funding_grant_id><funding_grant_id>R01 NS132344</funding_grant_id><funding_grant_id>R01NS132344</funding_grant_id><pubmed_authors>Gadd S</pubmed_authors><pubmed_authors>Clutter MR</pubmed_authors><pubmed_authors>Natsumeda M</pubmed_authors><pubmed_authors>Sasaki T</pubmed_authors><pubmed_authors>Uchida E</pubmed_authors><pubmed_authors>Abe K</pubmed_authors><pubmed_authors>Kaur S</pubmed_authors><pubmed_authors>Ishi Y</pubmed_authors><pubmed_authors>Watanabe J</pubmed_authors><pubmed_authors>Zhang Z</pubmed_authors><pubmed_authors>Becher OJ</pubmed_authors><pubmed_authors>Gullette MJ</pubmed_authors><pubmed_authors>Takata N</pubmed_authors><pubmed_authors>Mo Y</pubmed_authors><pubmed_authors>Hashizume R</pubmed_authors></additional><is_claimable>false</is_claimable><name>BET bromodomain inhibition potentiates radiosensitivity in models of H3K27-altered diffuse midline glioma.</name><description>Diffuse midline glioma (DMG) H3K27-altered is one of the most malignant childhood cancers.
