However, significant challenges that remain in top-down proteomics include the analysis of large proteoforms and the sensitivity required to examine proteoforms from minimal amounts of sample. To address these challenges, we have developed a new method termed "small-scale serial Size Exclusion Chromatography" (s3SEC), which incorporates a small-scale protein extraction (1 mg of tissue) and serial SEC without postfractionation sample handling, coupled with online high sensitivity capillary reversed-phase </pubmed_abstract><journal>Analytical chemistry</journal><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC11298573</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Small-Scale Serial Size Exclusion Chromatography (s3SEC) for High Sensitivity Top-Down Proteomics of Large Proteoforms.</pubmed_title><pmcid>PMC11298573</pmcid><funding_grant_id>R01 HL109810</funding_grant_id><funding_grant_id>T32 HL007936</funding_grant_id><funding_grant_id>R01 GM125085</funding_grant_id><funding_grant_id>DGE-1747503</funding_grant_id><funding_grant_id>S10 OD018475</funding_grant_id><funding_grant_id>R01 HL109810-06</funding_grant_id><funding_grant_id>T32 HL007936-21</funding_grant_id><funding_grant_id>R01 HL096971</funding_grant_id><pubmed_authors>Rogers HT</pubmed_authors><pubmed_authors>Larson EJ</pubmed_authors><pubmed_authors>Fischer MS</pubmed_authors><pubmed_authors>Rossler KJ</pubmed_authors><pubmed_authors>Ge Y</pubmed_authors><pubmed_authors>Wang D</pubmed_authors><pubmed_authors>Gao Z</pubmed_authors><pubmed_authors>Alpert AJ</pubmed_authors><pubmed_authors>Melby JA</pubmed_authors><pubmed_authors>Ehlers LE</pubmed_authors></additional><is_claimable>false</is_claimable>
Small-Scale Serial Size Exclusion Chromatography (s3SEC) for High Sensitivity Top-Down Proteomics of Large Proteoforms.

Top-down-mass spectrometry (MS)-based proteomics has emerged as a premier technology to examine proteins at the proteoform level, enabling characterization of genetic mutations, alternative splicing, and post-translational modifications.
