Infancy and childhood are crucial periods of rapid immunologic development, but how Tregs mediate immune responses at these earliest timepoints of human life is poorly understood. In this study, we compare blood and tissue (tonsil) Tregs across pediatric and adult subjects to investigate age-related differences in Treg biology. We observed increased FOXP3 expression and proportions of Tregs in tonsil compared with paired blood samples in children. Within tonsil, early life Tregs accumulated in extrafollicular regions with cellular interactions biased toward CD8+ T cells. Tonsil Tregs in both children and adults expressed tran</pubmed_abstract><journal>Journal of immunology (Baltimore, Md. : 1950)</journal><pubmed_title>Distinct Localization, Transcriptional Profiles, and Functionality in Early Life Tonsil Regulatory T Cells.</pubmed_title><pmcid>PMC11304551</pmcid><funding_grant_id>S10RR027050</funding_grant_id><funding_grant_id>P01 AI106697</funding_grant_id><funding_grant_id>UL1 TR001873</funding_grant_id><funding_grant_id>K23AI141686</funding_grant_id><funding_grant_id>S10 OD030282</funding_grant_id><funding_grant_id>UL1TR001873</funding_grant_id><funding_grant_id>S10OD020056</funding_grant_id><funding_grant_id>K23 AI141686</funding_grant_id><funding_grant_id>S10 OD020056</funding_grant_id><funding_grant_id>S10 RR027050</funding_grant_id><funding_grant_id>P01AI106697</funding_grant_id><funding_grant_id>U01AI100119</funding_grant_id><funding_grant_id>U01 AI100119</funding_grant_id><pubmed_authors>Verma S</pubmed_authors><pubmed_authors>Gray J</pubmed_authors><pubmed_authors>Bradley MC</pubmed_authors><pubmed_authors>Grunstein E</pubmed_authors><pubmed_authors>Pethe K</pubmed_authors><pubmed_authors>Dogra P</pubmed_authors><pubmed_authors>Maurrasse S</pubmed_authors><pubmed_authors>Waldman E</pubmed_authors><pubmed_authors>Connors TJ</pubmed_authors><pubmed_authors>Caron DP</pubmed_authors><pubmed_authors>Jang M</pubmed_authors><pubmed_authors>Farber DL</pubmed_authors></additional><is_claimable>false</is_claimable><name>Distinct Localization, Transcriptional Profiles, and Functionality in Early Life Tonsil Regulatory T Cells.</name><description>CD4+ regulatory T cells (Tregs) are key orchestrators of the immune system, fostering the establishment of protective immunity while preventing deleterious responses.
