We evaluated the efficacy of chimeric antigen receptor (CAR) T cell-mediated cancer cell lysis potential in the immune microenvironment of BCAA supplementation and deletion. BCAA supplementation increased cancer cell killing percentage, while accelerating BCAA catabolism and decreasing BCAA transporter decreased cancer cell lysis efficacy. We thus designed BCKDK engineering CAR T cells for the reprogramming of BCAA metabolism in the tumor microenvironment based on the genotype and phenotype modification. BCKDK overexpression (OE) in CAR-T cells significantly improved cancer cell lysis, while BCKDK knockout (KO) resulted in inferior lysis potential</pubmed_abstract><journal>Molecular therapy : the journal of the American Society of Gene Therapy</journal><pagination>3128-3144</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC11403223</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>BCKDK modification enhances the anticancer efficacy of CAR-T cells by reprogramming branched chain amino acid metabolism.</pubmed_title><pmcid>PMC11403223</pmcid><pubmed_authors>Yuan T</pubmed_authors><pubmed_authors>Shi L</pubmed_authors><pubmed_authors>Li C</pubmed_authors><pubmed_authors>Hao J</pubmed_authors><pubmed_authors>Han L</pubmed_authors><pubmed_authors>Zhou S</pubmed_authors><pubmed_authors>Han Y</pubmed_authors><pubmed_authors>Zhang L</pubmed_authors><pubmed_authors>Huang P</pubmed_authors><pubmed_authors>Gan R</pubmed_authors><pubmed_authors>Xin B</pubmed_authors><pubmed_authors>Zhu X</pubmed_authors><pubmed_authors>Yang Q</pubmed_authors><pubmed_authors>Guo C</pubmed_authors><pubmed_authors>Tu Y</pubmed_authors></additional><is_claimable>false</is_claimable><name>BCKDK modification enhances the anticancer efficacy of CAR-T cells by reprogramming branched chain amino acid metabolism.</name><description>Altered branched chain amino acids (BCAAs), including leucine, isoleucine, and valine, are frequently observed in patients with advanced cancer.
