Here, we report genome-wide profiles of primary LNETs' cis-regulatory elements by H3K27ac ChIP-seq with matching RNA-seq profiles. Analysis of these regulatory landscapes revealed three regulatory subtypes, independent of the typical/atypical classification. We identified unique differentiation signals that delineate each subtype. The "proneuronal" subtype emerges under the influence of ASCL1, SOX4, and TCF4 transcription factors, embodying a pronounced proneuronal signature. The "luminal-like" subtype is characterized by gain of acetylation at</pubmed_abstract><journal>Proceedings of the National Academy of Sciences of the United States of America</journal><pubmed_title>Enhancer landscape of lung neuroendocrine tumors reveals regulatory and developmental signatures with potential theranostic implications.</pubmed_title><pmcid>PMC11474083</pmcid><funding_grant_id>NA</funding_grant_id><pubmed_authors>Brim C</pubmed_authors><pubmed_authors>Izhar U</pubmed_authors><pubmed_authors>Hecht M</pubmed_authors><pubmed_authors>Antman I</pubmed_authors><pubmed_authors>Drier Y</pubmed_authors><pubmed_authors>Abu Akar F</pubmed_authors><pubmed_authors>Abu-Kamel S</pubmed_authors><pubmed_authors>Jumaa M</pubmed_authors><pubmed_authors>Wald O</pubmed_authors><pubmed_authors>Maimon O</pubmed_authors><pubmed_authors>Tzur T</pubmed_authors><pubmed_authors>Avniel-Polak S</pubmed_authors><pubmed_authors>Grozinsky-Glasberg S</pubmed_authors><pubmed_authors>Davis E</pubmed_authors><pubmed_authors>Maron Y</pubmed_authors><pubmed_authors>Atlan K</pubmed_authors><pubmed_authors>Saadoun T</pubmed_authors><pubmed_authors>Bel-Ange A</pubmed_authors></additional><is_claimable>false</is_claimable><name>Enhancer landscape of lung neuroendocrine tumors reveals regulatory and developmental signatures with potential theranostic implications.</name><description>Well-differentiated low-grade lung neuroendocrine tumors (lung carcinoids or LNETs) are histopathologically classified as typical and atypical LNETs, but each subtype is still heterogeneous at both the molecular level and its clinical manifestation.
