These mt-tRNAs have unusual sequence motifs and require post-transcriptional modifications to stabilize their fragile structures. However, whether a modification that stabilizes a wild-type (WT) mt-tRNA would also stabilize its pathogenic variants is unknown. Here we show that the N1-methylation of guanosine at position 9 (m1G9) of mt-Leu(UAA), while stabilizing the WT tRNA, has a destabilizing effect on variants associated with MELAS (mitochondrial myopathy, encephalopathy, lactic acidosis, and stroke-like episodes). This differential effect is further demonstrated, as removal of the m1G9 methylation, while damaging to the WT tR</pubmed_abstract><journal>Nature communications</journal><pubmed_title>Post-transcriptional methylation of mitochondrial-tRNA differentially contributes to mitochondrial pathology.</pubmed_title><pmcid>PMC11489592</pmcid><funding_grant_id>R35 GM149336</funding_grant_id><funding_grant_id>R35 GM134931</funding_grant_id><funding_grant_id>R01 AG078814</funding_grant_id><funding_grant_id>R01 GM123771</funding_grant_id><funding_grant_id>R01 CA259635</funding_grant_id><pubmed_authors>Maharjan S</pubmed_authors><pubmed_authors>Suzuki T</pubmed_authors><pubmed_authors>Christian T</pubmed_authors><pubmed_authors>Seifert E</pubmed_authors><pubmed_authors>Li NS</pubmed_authors><pubmed_authors>Gamper H</pubmed_authors><pubmed_authors>Hou YM</pubmed_authors><pubmed_authors>Wanunu M</pubmed_authors><pubmed_authors>Yamaki Y</pubmed_authors><pubmed_authors>Henley RY</pubmed_authors><pubmed_authors>Wallace DC</pubmed_authors><pubmed_authors>Piccirilli JA</pubmed_authors></additional><is_claimable>false</is_claimable>

Human mitochondrial tRNAs (mt-tRNAs), critical for mitochondrial biogenesis, are frequently associated with pathogenic mutations.
