Little is known about JCV pathogenesis, and no host factors required for cellular infection have been identified. Recently, we identified low-density lipoprotein receptor related protein 1 (Lrp1) as a host entry factor for two other bunyaviruses Rift Valley fever virus (RVFV) and Oropouche virus (OROV). Here, we assessed the role of Lrp1 in mediating JCV cellular infection of neurons. Both neuronal and non-neuronal immortalized cell lines deficient for Lrp1 displayed reduction in infection with JCV, and early stages of infection such as binding and internalization were impacted by lack of Lrp1. In primary rat neurons, Lrp1 was highly expressed, and the neurons were highly </pubmed_abstract><journal>bioRxiv : the preprint server for biology</journal><pagination>2024.11.06.622176</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC11580904</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Lrp1 facilitates infection of neurons by Jamestown Canyon virus.</pubmed_title><pmcid>PMC11580904</pmcid><funding_grant_id>R01 AI178378</funding_grant_id><funding_grant_id>UC7 AI180311</funding_grant_id><funding_grant_id>R01 AI161765</funding_grant_id><funding_grant_id>R56 AI171920</funding_grant_id><funding_grant_id>R01 AI169850</funding_grant_id><funding_grant_id>R01 NS101100</funding_grant_id><pubmed_authors>Sterling CE</pubmed_authors><pubmed_authors>Price DA</pubmed_authors><pubmed_authors>Frey ZD</pubmed_authors><pubmed_authors>Cui X</pubmed_authors><pubmed_authors>Connors KA</pubmed_authors><pubmed_authors>Brown G</pubmed_authors><pubmed_authors>Leung DW</pubmed_authors><pubmed_authors>Rush RE</pubmed_authors><pubmed_authors>Ganaie S</pubmed_authors><pubmed_authors>Amarasinghe GK</pubmed_authors><pubmed_authors>Wills ZP</pubmed_authors><pubmed_authors>Hartman AL</pubmed_authors><pubmed_authors>Fatma F</pubmed_authors><pubmed_authors>Schwarz MM</pubmed_authors></additional><is_claimable>false</is_claimable><name>Lrp1 facilitates infection of neurons by Jamestown Canyon virus.</name><description>Jamestown Canyon virus (JCV) is a bunyavirus and arbovirus responsible for neuroinvasive disease in the United States.
