However, the complexity of TME in AITL progression is poorly understood. We performed single-cell RNA-Seq (scRNA-seq) and imaging mass cytometry (IMC) analysis to compare the cellular composition and spatial architecture between relapsed/refractory AITL (RR-AITL) and newly diagnosed AITL (ND-AITL). Our results showed that the malignant T follicular helper (Tfh) cells showed significantly increased proliferation driven by transcriptional activation of YY1 in RR-AITL, which is markedly associated with the poor prognosis of AITL patients. The CD8&lt;sup>+&lt;/sup> T cell proportion and cytotoxicity decreased in RR-AITL TME, resulting from</pubmed_abstract><journal>Blood cancer journal</journal><pubmed_title>Single-cell transcriptomic and spatial analysis reveal the immunosuppressive microenvironment in relapsed/refractory angioimmunoblastic T-cell lymphoma.</pubmed_title><pmcid>PMC11655871</pmcid><funding_grant_id>91959113</funding_grant_id><funding_grant_id>BK20210530</funding_grant_id><funding_grant_id>82370193</funding_grant_id><funding_grant_id>32200590</funding_grant_id><funding_grant_id>BK20211376</funding_grant_id><funding_grant_id>82170185</funding_grant_id><funding_grant_id>82372897</funding_grant_id><funding_grant_id>81972358</funding_grant_id><pubmed_authors>Cao L</pubmed_authors><pubmed_authors>Yu L</pubmed_authors><pubmed_authors>Wu L</pubmed_authors><pubmed_authors>Lu X</pubmed_authors><pubmed_authors>Wang C</pubmed_authors><pubmed_authors>Ren Y</pubmed_authors><pubmed_authors>Hu X</pubmed_authors><pubmed_authors>Liu Q</pubmed_authors><pubmed_authors>Shi W</pubmed_authors><pubmed_authors>Zhang L</pubmed_authors><pubmed_authors>Lin Z</pubmed_authors><pubmed_authors>Huang R</pubmed_authors><pubmed_authors>Zhang R</pubmed_authors><pubmed_authors>Shi Y</pubmed_authors><pubmed_authors>Leng J</pubmed_authors><pubmed_authors>Jin H</pubmed_authors><pubmed_authors>Wang Q</pubmed_authors><pubmed_authors>Fan L</pubmed_authors><pubmed_authors>Wu R</pubmed_authors><pubmed_authors>Wu W</pubmed_authors><pubmed_authors>Huang B</pubmed_authors><pubmed_authors>Chen R</pubmed_authors><pubmed_authors>Li J</pubmed_authors><pubmed_authors>Lu C</pubmed_authors><pubmed_authors>Xia P</pubmed_authors><pubmed_authors>Zhou W</pubmed_authors><pubmed_authors>Li K</pubmed_authors><pubmed_authors>Li N</pubmed_authors><pubmed_authors>Zhu M</pubmed_authors><pubmed_authors>Zhong S</pubmed_authors><pubmed_authors>Yao D</pubmed_authors><pubmed_authors>Cai Y</pubmed_authors><pubmed_authors>Lv S</pubmed_authors></additional><is_claimable>false</is_claimable><name>Single-cell transcriptomic and spatial analysis reveal the immunosuppressive microenvironment in relapsed/refractory angioimmunoblastic T-cell lymphoma.</name><description>Angioimmunoblastic T-cell lymphoma (AITL) is a kind of aggressive T-cell lymphoma with significant enrichment of non-malignant tumor microenvironment (TME) cells.
