Using layer-by-layer mutant design and femtosecond spectroscopy, we have successfully determined the rates of electron transfer and proton transfer, driving force, and reorganization energy for nine elementary steps involved in the initial photoreduction of class II <i>Arabidopsis thaliana</i> photolyase (AtPL), thereby constructing the photoreduction network specific to class II PLs. Several dynamic features have been revealed including a slow-rise (172 ps) and</pubmed_abstract><journal>Proceedings of the National Academy of Sciences of the United States of America</journal><pubmed_title>Elucidation of a distinct photoreduction pathway in class II &lt;i&gt;Arabidopsis thaliana&lt;/i&gt; photolyase.</pubmed_title><pmcid>PMC11725880</pmcid><funding_grant_id>31971233</funding_grant_id><funding_grant_id>12004246</funding_grant_id><funding_grant_id>22433003</funding_grant_id><funding_grant_id>2020YFA0509700</funding_grant_id><pubmed_authors>Ding B</pubmed_authors><pubmed_authors>Chen Z</pubmed_authors><pubmed_authors>Wang B</pubmed_authors><pubmed_authors>Kang XW</pubmed_authors><pubmed_authors>Zhong D</pubmed_authors><pubmed_authors>Tang S</pubmed_authors><pubmed_authors>Zou S</pubmed_authors><pubmed_authors>Zhou Y</pubmed_authors><pubmed_authors>Zhou Z</pubmed_authors></additional><is_claimable>false</is_claimable>

Class II photolyases (PLs) are a distant subclade in the photolyase/cryptochrome superfamily, displaying a unique Trp-Tyr tetrad for photoreduction and exhibiting a lower quantum yield (QY) of DNA repair (49%) than class I photolyases (82%) [M.
