In this study, it is shown that eEF2K is highly expressed in TNBC and is associated with poor prognosis. In vitro, in vivo, and patient-derived organoid experiments demonstrate that knockdown of eEF2K significantly impedes progression of TNBC. Proteomic analysis and confirmation experiments reveal that eEF2K positively regulates the mRNA and protein expressions of sex-determining region Y-box 8 (SOX8). Mechanistically, eEF2K binds to and phosphorylates aurora kinase A (AURKA) at S391, a newly identified phosphorylation site critical for maintaining AURKA prote</pubmed_abstract><journal>Advanced science (Weinheim, Baden-Wurttemberg, Germany)</journal><pubmed_title>Identification of a Novel Substrate for eEF2K and the AURKA-SOX8 as the Related Pathway in TNBC.</pubmed_title><pmcid>PMC11984844</pmcid><funding_grant_id>81972480</funding_grant_id><funding_grant_id>82373064</funding_grant_id><funding_grant_id>1053320221775</funding_grant_id><funding_grant_id>22477093</funding_grant_id><funding_grant_id>22177086</funding_grant_id><pubmed_authors>Yang J</pubmed_authors><pubmed_authors>Gong R</pubmed_authors><pubmed_authors>Zhong C</pubmed_authors><pubmed_authors>Jiang S</pubmed_authors><pubmed_authors>Wan X</pubmed_authors><pubmed_authors>He L</pubmed_authors><pubmed_authors>Yi W</pubmed_authors><pubmed_authors>Cheng Y</pubmed_authors><pubmed_authors>Zhao X</pubmed_authors><pubmed_authors>Li Y</pubmed_authors><pubmed_authors>Ye N</pubmed_authors><pubmed_authors>Chen Z</pubmed_authors><pubmed_authors>Cao S</pubmed_authors><pubmed_authors>Tian S</pubmed_authors><pubmed_authors>Zhu R</pubmed_authors><pubmed_authors>Shan T</pubmed_authors></additional><is_claimable>false</is_claimable><name>Identification of a Novel Substrate for eEF2K and the AURKA-SOX8 as the Related Pathway in TNBC.</name><description>Eukaryotic elongation factor 2 kinase (eEF2K) has been considered as a putative target for cancer therapy; however, the underlying mechanisms of eEF2K in triple-negative breast cancer (TNBC) progression remain to be fully elucidated.
