Thus, to address the question of whether alphabeta T cells could recognize MHC molecules independently of bound ligands, we studied human and mouse Hfe interactions with T lymphocytes. We provide formal evidence of direct cytolytic recognition of human Hfe by mouse alphabeta T cell receptors (TCR) in HLA-A*0201 transgenic mice and that this interaction results in ZAP-70 phosphorylation. Furthermore, direct recognition of mouse Hfe molecules by cytotoxic T lymphocytes (CTLs) was demonstrated in DBA/2 Hfe knockout mice. These CTLs express predominantly two T cell antigen receptor alpha variable gene segments (AV6.1 and AV6.</pubmed_abstract><journal>Proceedings of the National Academy of Sciences of the United States of America</journal><pagination>12855-60</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC1200262</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Direct recognition by alphabeta cytolytic T cells of Hfe, a MHC class Ib molecule without antigen-presenting function.</pubmed_title><pmcid>PMC1200262</pmcid><pubmed_authors>Charneau P</pubmed_authors><pubmed_authors>Fazilleau N</pubmed_authors><pubmed_authors>Pascolo S</pubmed_authors><pubmed_authors>Coppin H</pubmed_authors><pubmed_authors>Rohrlich PS</pubmed_authors><pubmed_authors>Lemonnier FA</pubmed_authors><pubmed_authors>Roth MP</pubmed_authors><pubmed_authors>Laham N</pubmed_authors><pubmed_authors>Michel F</pubmed_authors><pubmed_authors>Ginhoux F</pubmed_authors><pubmed_authors>Nato F</pubmed_authors><pubmed_authors>Danos O</pubmed_authors><pubmed_authors>Cochet M</pubmed_authors><pubmed_authors>Acuto O</pubmed_authors><pubmed_authors>Ehrlich R</pubmed_authors><pubmed_authors>Kanellopoulos J</pubmed_authors><pubmed_authors>Firat H</pubmed_authors></additional><is_claimable>false</is_claimable><name>Direct recognition by alphabeta cytolytic T cells of Hfe, a MHC class Ib molecule without antigen-presenting function.</name><description>Crystallographic analysis of human Hfe has documented an overall structure similar to classical (class Ia) MHC molecules with a peptide binding groove deprived of ligand.
