Bioleaching offers a promising alternative to conventional hydrometallurgical methods for REE extraction from low-grade ores. However, exploiting this potential remains challenging due to large gaps in our understanding of the genetics involved, and inadequate biological tools to address them. We generated a highly non-redundant whole-genome knockout collection for the bioleaching microbe Gluconobacter oxydans B58, reducing redundancy by 85% compared to the previous best collection. This new collection was directly screened for bioleaching neodymium from a synthetic monazite powder, identifying 89 genes important for bioleaching, 68 of which have not previously been</pubmed_abstract><journal>Communications biology</journal><pubmed_title>Direct genome-scale screening of Gluconobacter oxydans B58 for rare earth element bioleaching.</pubmed_title><pmcid>PMC12041372</pmcid><funding_grant_id>2435259</funding_grant_id><funding_grant_id>DE-AR0001341</funding_grant_id><pubmed_authors>Pian B</pubmed_authors><pubmed_authors>Schmitz A</pubmed_authors><pubmed_authors>Wu M</pubmed_authors><pubmed_authors>Medin SA</pubmed_authors><pubmed_authors>Holycross M</pubmed_authors><pubmed_authors>Reid MC</pubmed_authors><pubmed_authors>Andrade M</pubmed_authors><pubmed_authors>Balta JB</pubmed_authors><pubmed_authors>Gazel E</pubmed_authors><pubmed_authors>Marecos S</pubmed_authors><pubmed_authors>Barstow B</pubmed_authors></additional><is_claimable>false</is_claimable><name>Direct genome-scale screening of Gluconobacter oxydans B58 for rare earth element bioleaching.</name><description>The transition to a sustainable energy economy will require an enormous increase in the supply of rare earth elements (REEs).
