However, direct pharmacological targeting of CTNNB1 has remained challenging. Here, we have performed a screen with a library of cysteine-reactive covalent ligands to identify the monovalent degrader EN83 that depletes CTNNB1 in a ubiquitin-proteasome-dependent manner. We show that EN83 directly and covalently targets CTNNB1 three cysteines C466, C520, and C619, leading to destabilization and degradation of CTNNB1. Through structural optimization, we generate a highly potent and relatively selective destabilizing degrader that acts through the targeting of only C619 on CTNNB</pubmed_abstract><journal>Journal of the American Chemical Society</journal><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC12102902</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Covalent Degrader of the Oncogenic Transcription Factor β-Catenin.</pubmed_title><pmcid>PMC12102902</pmcid><funding_grant_id>S10 OD024998</funding_grant_id><funding_grant_id>R01 CA240981</funding_grant_id><funding_grant_id>R35 CA263814</funding_grant_id><funding_grant_id>R35CA263814</funding_grant_id><funding_grant_id>R01CA240981</funding_grant_id><pubmed_authors>Forte N</pubmed_authors><pubmed_authors>Nomura DK</pubmed_authors><pubmed_authors>Altamirano Poblano BE</pubmed_authors><pubmed_authors>Huang OW</pubmed_authors><pubmed_authors>Hatcher J</pubmed_authors><pubmed_authors>Wang Y</pubmed_authors><pubmed_authors>Wertz IE</pubmed_authors><pubmed_authors>Gowans FA</pubmed_authors></additional><is_claimable>false</is_claimable><name>Covalent Degrader of the Oncogenic Transcription Factor β-Catenin.</name><description>β-catenin (CTNNB1) is an oncogenic transcription factor that is important in cell-cell adhesion and transcription of cell proliferation and survival genes that drive the pathogenesis of many different types of cancers.
