Recent data in mouse and human have revealed non-T, non-ILC3 cell populations with antigen presenting features that express RORγt. This study maps the presence of RORγt cells with dendritic cell (DC) features in human adult and fetal lymphoid tissues.
Methods
By combining multicolor flow cytometry, RNAseq of peripheral blood cells, analysis of lymph node scRNAseq datasets, and microscopic analysis on human tissue sections and single-cell suspensions, this study maps the presence of RORγt cells with DC features in human tissues.
Results
RORγt-DCs are found in human lymphoid</pubmed_abstract><journal>Frontiers in immunology</journal><pagination>1527499</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC12162286</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Mapping of RORγt+ dendritic cells in human tissues establishes their preferential niche in adult lymph nodes.</pubmed_title><pmcid>PMC12162286</pmcid><pubmed_authors>Bianchetto-Aguilera FM</pubmed_authors><pubmed_authors>Monti M</pubmed_authors><pubmed_authors>Missale F</pubmed_authors><pubmed_authors>Bugatti M</pubmed_authors><pubmed_authors>Antonova AU</pubmed_authors><pubmed_authors>Lonardi S</pubmed_authors><pubmed_authors>Ferrari G</pubmed_authors><pubmed_authors>Colonna M</pubmed_authors><pubmed_authors>Lorenzi L</pubmed_authors><pubmed_authors>Savoldi V</pubmed_authors><pubmed_authors>Cassatella MA</pubmed_authors><pubmed_authors>Di Matteo A</pubmed_authors><pubmed_authors>Zini S</pubmed_authors><pubmed_authors>Ghigna C</pubmed_authors><pubmed_authors>Cella M</pubmed_authors><pubmed_authors>Benedetti M</pubmed_authors><pubmed_authors>Facchetti F</pubmed_authors><pubmed_authors>Wu S</pubmed_authors><pubmed_authors>Agostini V</pubmed_authors><pubmed_authors>Picinoli S</pubmed_authors><pubmed_authors>Vermi W</pubmed_authors><pubmed_authors>Chiarini M</pubmed_authors><pubmed_authors>Liu YA</pubmed_authors><pubmed_authors>Moratto D</pubmed_authors></additional><is_claimable>false</is_claimable>

Background
The retinoic-acid-receptor-related orphan receptor gamma t (RORγt) isoform is required for the development of lymphoid organs, T-helper 17 cells (Th17), and innate lymphoid cells (ILC3s).
