Here, we used single cell (sc)-RNA-Seq analysis of UM-hepatic stellate cell (HSC) co-cultures to demonstrate that HSCs enriched for UM cell states that expressed genes implicated in cell survival, metabolic reprogramming and angiogenesis. A lead candidate driver of HSC reprogramming was the TGF-β family member GDF15, which was associated with a metastatic UM phenotype. Silencing of BAP1 in UM cells led to increased GDF15 expression and accumulation of H3K27ac marks at the GDF15 promoter. Treatment of HSCs with GDF15 led to increased expression extracellular matrix proteins, inflammatory cytokines </pubmed_abstract><journal>bioRxiv : the preprint server for biology</journal><pagination>2025.05.07.652654</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC12247901</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>GDF15 reprograms the microenvironment to drive the development of uveal melanoma liver metastases.</pubmed_title><pmcid>PMC12247901</pmcid><funding_grant_id>P30 CA247796</funding_grant_id><funding_grant_id>R01 CA256193</funding_grant_id><funding_grant_id>P30 CA142543</funding_grant_id><funding_grant_id>P30 CA076292</funding_grant_id><funding_grant_id>P30 EY030413</funding_grant_id><pubmed_authors>Kurtenbach S</pubmed_authors><pubmed_authors>Chen YA</pubmed_authors><pubmed_authors>Dollar JJ</pubmed_authors><pubmed_authors>Siddarajappa N</pubmed_authors><pubmed_authors>Nguyen T</pubmed_authors><pubmed_authors>Harbour JW</pubmed_authors><pubmed_authors>Smalley KSM</pubmed_authors><pubmed_authors>Licht JD</pubmed_authors><pubmed_authors>Park JS</pubmed_authors><pubmed_authors>Kuznetsoff JN</pubmed_authors><pubmed_authors>Adams DJ</pubmed_authors><pubmed_authors>Sriramareddy SN</pubmed_authors><pubmed_authors>Correa ZM</pubmed_authors></additional><is_claimable>false</is_claimable><name>GDF15 reprograms the microenvironment to drive the development of uveal melanoma liver metastases.</name><description>Uveal melanoma (UM) results in fatal liver metastasis, yet little is known about the interactions between UM and host cells in the tumor microenvironment that promote this distinctive proclivity.
