This study aimed to clarify the mechanisms connecting the LD aging and herniation, particularly focusing on cellular senescence and molecular alterations in the nucleus pulposus (NP). We performed a detailed analysis of NP samples from a diverse cohort, including individuals of varying ages and those with diagnosed LD herniation. Our methodology combined histological assessments with single-nucleus RNA sequencing to identify phenotypic and molecular changes related to NP aging and herniation. We discovered that cellular senescence and a decrease in nucleus pulposus progenitor cells (NPPCs) are central to both processes. Additionally, we found an age-related increase in NFAT1 expression </pubmed_abstract><journal>Protein &amp; cell</journal><pubmed_title>Single-nucleus transcriptomics decodes the link between aging and lumbar disc herniation.</pubmed_title><pmcid>PMC12342153</pmcid><funding_grant_id>JYY2023-13</funding_grant_id><funding_grant_id>82322025</funding_grant_id><funding_grant_id>ZJAD-2024001</funding_grant_id><funding_grant_id>82361148131</funding_grant_id><funding_grant_id>2022YFA1103700</funding_grant_id><funding_grant_id>YSBR-012</funding_grant_id><funding_grant_id>2022YFA1103800</funding_grant_id><funding_grant_id>STI2030-Major Projects-2021ZD0202400</funding_grant_id><funding_grant_id>YSBR-076</funding_grant_id><funding_grant_id>XDA0460403</funding_grant_id><funding_grant_id>82361148130</funding_grant_id><funding_grant_id>2021-1045</funding_grant_id><funding_grant_id>82471586</funding_grant_id><funding_grant_id>82271600</funding_grant_id><funding_grant_id>Z230011</funding_grant_id><funding_grant_id>2020YFA0804000</funding_grant_id><funding_grant_id>8231101626</funding_grant_id><funding_grant_id>2022083</funding_grant_id><funding_grant_id>HYZHXMH01012</funding_grant_id><funding_grant_id>C2406001</funding_grant_id><funding_grant_id>JQ24044</funding_grant_id><funding_grant_id>82422031</funding_grant_id><funding_grant_id>82488301</funding_grant_id><funding_grant_id>12300927</funding_grant_id><funding_grant_id>XDC0200000</funding_grant_id><funding_grant_id>82125011</funding_grant_id><funding_grant_id>CAS-WX2022SDC-XK14</funding_grant_id><funding_grant_id>32121001</funding_grant_id><funding_grant_id>32341001</funding_grant_id><funding_grant_id>92468303</funding_grant_id><funding_grant_id>92168201</funding_grant_id><funding_grant_id>2023IOZ0102</funding_grant_id><funding_grant_id>2023IOZ0202</funding_grant_id><funding_grant_id>2024IOZ0103</funding_grant_id><funding_grant_id>82192863</funding_grant_id><funding_grant_id>Z240018</funding_grant_id><funding_grant_id>2021QNRC001</funding_grant_id><funding_grant_id>BPHR202203105</funding_grant_id><funding_grant_id>82330044</funding_grant_id><pubmed_authors>Li J</pubmed_authors><pubmed_authors>Ma S</pubmed_authors><pubmed_authors>Wang A</pubmed_authors><pubmed_authors>Li C</pubmed_authors><pubmed_authors>Lei J</pubmed_authors><pubmed_authors>Liu GH</pubmed_authors><pubmed_authors>Qu J</pubmed_authors><pubmed_authors>Yang C</pubmed_authors><pubmed_authors>Wang M</pubmed_authors><pubmed_authors>Zhang W</pubmed_authors><pubmed_authors>He Z</pubmed_authors><pubmed_authors>Wang S</pubmed_authors><pubmed_authors>Sun S</pubmed_authors><pubmed_authors>Yu Z</pubmed_authors><pubmed_authors>Liu F</pubmed_authors><pubmed_authors>Yu Y</pubmed_authors></additional><is_claimable>false</is_claimable><name>Single-nucleus transcriptomics decodes the link between aging and lumbar disc herniation.</name><description>Lumbar disc (LD) herniation and aging are prevalent conditions that can result in substantial morbidity.
