Here we show that tissue proteomics provide critical insights not captured by tissue gene expression or blood proteomics information by performing whole-genome and RNA sequencing and proteomics analysis of human kidney samples (n = 337), and we generated a publicly available database. Via Bayesian co-localization and Mendelian randomization analyses of kidney protein quantitative trait loci and 36 CKM genome-wide association studies, we prioritized 89 proteins for CKM traits. We prioritized relationships that could underlie the interconnectedness of CKM traits and discovered multiple and targetable mechanisms for C</pubmed_abstract><journal>Nature medicine</journal><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC12435990</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>The proteogenomic landscape of the human kidney and implications for cardio-kidney-metabolic health.</pubmed_title><pmcid>PMC12435990</pmcid><funding_grant_id>R01 DK087635</funding_grant_id><funding_grant_id>R01 DK123459</funding_grant_id><funding_grant_id>R01 DK076077</funding_grant_id><funding_grant_id>R01 DK105821</funding_grant_id><pubmed_authors>Liu H</pubmed_authors><pubmed_authors>Vitale S</pubmed_authors><pubmed_authors>Niewczas MA</pubmed_authors><pubmed_authors>Li C</pubmed_authors><pubmed_authors>Fadista J</pubmed_authors><pubmed_authors>Susztak K</pubmed_authors><pubmed_authors>Zeng L</pubmed_authors><pubmed_authors>Nguyen T</pubmed_authors><pubmed_authors>Olsson N</pubmed_authors><pubmed_authors>Abedini A</pubmed_authors><pubmed_authors>Mohandes S</pubmed_authors><pubmed_authors>Levinsohn J</pubmed_authors><pubmed_authors>McAllister FE</pubmed_authors><pubmed_authors>Vassalotti A</pubmed_authors><pubmed_authors>Shungin D</pubmed_authors><pubmed_authors>Karihaloo A</pubmed_authors><pubmed_authors>Hirohama D</pubmed_authors><pubmed_authors>Ha E</pubmed_authors></additional><is_claimable>false</is_claimable><name>The proteogenomic landscape of the human kidney and implications for cardio-kidney-metabolic health.</name><description>Nearly one-third of the global population is affected by cardio-kidney-metabolic (CKM) diseases; however, the molecular mechanisms underlying CKM diseases are poorly understood.
