We previously reported selective expression of CD37 on malignant T cells in a subset of TCL. Herein, we demonstrate CAR-37 T cells specifically target CD37-positive TCL in part by activating the intrinsic apoptotic pathway. To maximize therapeutic index, we identified selective/targetable BH3 dependences in individual TCL models and combined with CAR-37 T cells. We show that BH3 mimetics do not alter CD37 antigen binding capacity on TCL and have minimal effects on CAR-37 T-cell phenotype or function. In TCL models with dependence on MCL-1, combining CAR-37 T cells and the MCL-1 inhibitor AZD5991 increases anti-TCL response </pubmed_abstract><journal>Leukemia</journal><pubmed_title>Combining MCL-1 inhibition and CD37-directed chimeric antigen receptor T cells as an effective strategy to target T-cell lymphoma.</pubmed_title><pmcid>PMC12463675</pmcid><funding_grant_id>R01 CA249185</funding_grant_id><funding_grant_id>R01 CA249062</funding_grant_id><funding_grant_id>R35 CA231958</funding_grant_id><funding_grant_id>R35 CA210030</funding_grant_id><funding_grant_id>P01 CA233412</funding_grant_id><pubmed_authors>Stegmaier K</pubmed_authors><pubmed_authors>Lohr JG</pubmed_authors><pubmed_authors>Liu J</pubmed_authors><pubmed_authors>Letai A</pubmed_authors><pubmed_authors>Wiggers CRM</pubmed_authors><pubmed_authors>Korell F</pubmed_authors><pubmed_authors>Wu G</pubmed_authors><pubmed_authors>Hegel J</pubmed_authors><pubmed_authors>Weinstock DM</pubmed_authors><pubmed_authors>B AT</pubmed_authors><pubmed_authors>Eisenbies Z</pubmed_authors><pubmed_authors>Silva H</pubmed_authors><pubmed_authors>Powers F</pubmed_authors><pubmed_authors>Scarfo I</pubmed_authors><pubmed_authors>Heavican-Foral TB</pubmed_authors><pubmed_authors>Knoechel B</pubmed_authors><pubmed_authors>Maus MV</pubmed_authors><pubmed_authors>Kulp S</pubmed_authors></additional><is_claimable>false</is_claimable><name>Combining MCL-1 inhibition and CD37-directed chimeric antigen receptor T cells as an effective strategy to target T-cell lymphoma.</name><description>Chimeric antigen receptor (CAR) T cell therapy has not yet been realized for T-cell lymphomas (TCL), partially due to challenges in identifying tumor-specific antigens.
