Effective therapeutic strategies are limited, and resistance to treatment is a major obstacle. Myeloid cell leukemia (MCL)-1 and B-cell leukemia (BCL)-xL are antiapoptotic B-cell lymphoma 2 (Bcl-2) family proteins that block cell-intrinsic apoptosis through interactions on the mitochondrial outer membrane which contribute to therapeutic resistance. We investigated whether B-cell homology domain3 profiles were consistent between intra-patient fresh tumor sample, patient-derived cells, and patient-derived xenografts (PDX) by B-cell homology domain-3 profiling; we observed striking consistency which enabled cross-model comparisons. Next, we co-targeted BCL-xl and MCL-1 and noted that</pubmed_abstract><journal>Molecular cancer therapeutics</journal><pubmed_title>Co-Targeting BCL-xL with MCL-1 Induces Lethal Mitochondrial Dysfunction in Diffuse Mesothelioma.</pubmed_title><pmcid>PMC12475887</pmcid><funding_grant_id>R37 CA289419</funding_grant_id><funding_grant_id>UM1 HG006348</funding_grant_id><funding_grant_id>R01 DK114356</funding_grant_id><funding_grant_id>R03 CA252685-01</funding_grant_id><funding_grant_id>R21 CA256466</funding_grant_id><funding_grant_id>U01 CA214195</funding_grant_id><funding_grant_id>R01 CA290173</funding_grant_id><funding_grant_id>S10 RR024574</funding_grant_id><funding_grant_id>R03 CA252685</funding_grant_id><funding_grant_id>U24 OH009077</funding_grant_id><funding_grant_id>P30 CA125123</funding_grant_id><funding_grant_id>R01 HL130249</funding_grant_id><pubmed_authors>Vilchis M</pubmed_authors><pubmed_authors>Zhu Y</pubmed_authors><pubmed_authors>Ripley RT</pubmed_authors><pubmed_authors>Hilsenbeck SG</pubmed_authors><pubmed_authors>Surman DR</pubmed_authors><pubmed_authors>Aggison JC</pubmed_authors><pubmed_authors>Shen YH</pubmed_authors><pubmed_authors>Li N</pubmed_authors><pubmed_authors>Ramineni M</pubmed_authors><pubmed_authors>Li Y</pubmed_authors><pubmed_authors>Medina CG</pubmed_authors><pubmed_authors>Wu S</pubmed_authors><pubmed_authors>Chen X</pubmed_authors><pubmed_authors>Dobrolecki LE</pubmed_authors><pubmed_authors>Xu Y</pubmed_authors></additional><is_claimable>false</is_claimable><name>Co-Targeting BCL-xL with MCL-1 Induces Lethal Mitochondrial Dysfunction in Diffuse Mesothelioma.</name><description>Diffuse mesothelioma is a rare but highly aggressive and treatment-resistant neoplasm with low survival rates.
