Food processing techniques can modify the food protein properties by altering their biophysical characteristics and allergen activity. This study examines the phenylglyoxal-based chemical modification of the cashew nut allergen Ana o 3. Immunoassays with multiple antibodies demonstrate reduced recognition of phenylglyoxal-modified Ana o 3. Mass spectrometry identified multiple Ana o 3 modification sites, including arginine 41, 54, 85, and 111. Circular dichroism, biochemical assays, and molecular simulation indicate that the modifications resulted in minimal protein structure alteration. The research presented here provides insight into Ana o 3 surface chemistry and structure, and it could be applied </pubmed_abstract><journal>Journal of agricultural and food chemistry</journal><pubmed_title>Phenylglyoxal-induced Ana o 3 Modification Reduces Antibody Binding with Minimal Alteration in Protein Structure.</pubmed_title><pmcid>PMC12492407</pmcid><funding_grant_id>0201-88888-003-000D</funding_grant_id><funding_grant_id>0201-88888-002-000D</funding_grant_id><funding_grant_id>2022-70001-37580</funding_grant_id><funding_grant_id>6054-43440-052-000-D</funding_grant_id><pubmed_authors>Rao Q</pubmed_authors><pubmed_authors>Dupre RA</pubmed_authors><pubmed_authors>Vuong TT</pubmed_authors><pubmed_authors>Mattison CP</pubmed_authors><pubmed_authors>Weigle AT</pubmed_authors><pubmed_authors>Smith B</pubmed_authors><pubmed_authors>Brown CN</pubmed_authors><pubmed_authors>Boue SM</pubmed_authors><pubmed_authors>Ebmeier CC</pubmed_authors><pubmed_authors>Chou YJ</pubmed_authors></additional><is_claimable>false</is_claimable><name>Phenylglyoxal-induced Ana o 3 Modification Reduces Antibody Binding with Minimal Alteration in Protein Structure.</name><description>The nutritional benefits of nut consumption are complicated by the presence of allergens.
