This study aims to investigate the clinical and biological features of CLL with t(14;19)(q32;q13) (tCLL) involving the BCL3 gene. Patients with tCLL were younger and more commonly presented unmutated IGHV gene, subset #8 stereotypy, trisomy of chromosome 12, and complex karyotype than other patients without t(14;19) (oCLL). The presence of t(14;19) was associated with a shorter time to treatment and overall survival compared to oCLL. Gene expression analysis revealed a unique transcriptome profile in tCLL, characterized by the upregulation of BCL3 and the activation of B-cell receptor, PI3K-</pubmed_abstract><journal>Leukemia</journal><pubmed_title>Clinical and transcriptomic characterization of patients with chronic lymphocytic leukemia harboring t(14;19): an ERIC study.</pubmed_title><pmcid>PMC12634426</pmcid><funding_grant_id>Be_CL3VER</funding_grant_id><funding_grant_id>IG-25024</funding_grant_id><funding_grant_id>21198</funding_grant_id><pubmed_authors>Cuneo A</pubmed_authors><pubmed_authors>Davis Z</pubmed_authors><pubmed_authors>Puiggros A</pubmed_authors><pubmed_authors>Trentin L</pubmed_authors><pubmed_authors>Kater A</pubmed_authors><pubmed_authors>Orsi S</pubmed_authors><pubmed_authors>Roncaglia E</pubmed_authors><pubmed_authors>Baran-Marszak F</pubmed_authors><pubmed_authors>Yi S</pubmed_authors><pubmed_authors>Serafin A</pubmed_authors><pubmed_authors>Woyach JA</pubmed_authors><pubmed_authors>Haferlach C</pubmed_authors><pubmed_authors>Terol MJ</pubmed_authors><pubmed_authors>Scarfo L</pubmed_authors><pubmed_authors>Doubek M</pubmed_authors><pubmed_authors>Bortoluzzi S</pubmed_authors><pubmed_authors>Miller C</pubmed_authors><pubmed_authors>Ghia P</pubmed_authors><pubmed_authors>Cui C</pubmed_authors><pubmed_authors>Rigolin GM</pubmed_authors><pubmed_authors>Stamatopoulos K</pubmed_authors><pubmed_authors>Abrisqueta P</pubmed_authors><pubmed_authors>Martines A</pubmed_authors><pubmed_authors>Chatzikonstantinou T</pubmed_authors><pubmed_authors>Panagiotidis P</pubmed_authors><pubmed_authors>Kalashnikova O</pubmed_authors><pubmed_authors>Cellini A</pubmed_authors><pubmed_authors>Espinet B</pubmed_authors><pubmed_authors>Valiente A</pubmed_authors><pubmed_authors>Bonaldi L</pubmed_authors><pubmed_authors>Kreuzer KA</pubmed_authors><pubmed_authors>Eichhorst B</pubmed_authors><pubmed_authors>Oscier D</pubmed_authors><pubmed_authors>Furstenau M</pubmed_authors><pubmed_authors>Calabretto G</pubmed_authors><pubmed_authors>Athanasiadou A</pubmed_authors><pubmed_authors>Frezzato F</pubmed_authors><pubmed_authors>Nguyen-Khac F</pubmed_authors><pubmed_authors>Visentin A</pubmed_authors><pubmed_authors>Gaffo E</pubmed_authors><pubmed_authors>Mauro FR</pubmed_authors><pubmed_authors>Bosch F</pubmed_authors><pubmed_authors>Panagiotis B</pubmed_authors><pubmed_authors>Tausch E</pubmed_authors><pubmed_authors>Ruocco V</pubmed_authors><pubmed_authors>Angotzi F</pubmed_authors><pubmed_authors>Rogers KA</pubmed_authors><pubmed_authors>Plevova K</pubmed_authors></additional><is_claimable>false</is_claimable><name>Clinical and transcriptomic characterization of patients with chronic lymphocytic leukemia harboring t(14;19): an ERIC study.</name><description>In chronic lymphocytic leukemia (CLL), the role of complex karyotype (CK) for prognostic stratification remains a topic of debate, and the impact of specific cytogenetic abnormalities is still unclear.
