Epigenetic mechanisms govern cellular glucose sensing and GSIS by β cells, but they remain incompletely defined. Here, we found that BAF60a functions as a chromatin regulator that sustains biphasic GSIS and preserves β cell function under metabolic stress conditions. BAF60a was downregulated in β cells from obese and diabetic mice, monkeys, and humans. β cell-specific inactivation of BAF60a in adult mice impaired GSIS, leading to hyperglycemia and glucose intolerance. Conversely, restoring BAF60a expression improved β cell function and systemic glucose homeostasis. Mechanistically, BAF60a physically interacted with Nkx6.1 to selectively modulate chromatin accessibility and transcriptional </pubmed_abstract><journal>The Journal of clinical investigation</journal><pubmed_title>BAF60a-dependent chromatin remodeling preserves β cell function and contributes to the therapeutic benefits of GLP-1R agonists.</pubmed_title><pmcid>PMC12646663</pmcid><funding_grant_id>2023ZKZD17</funding_grant_id><funding_grant_id>LHDMD24H030001,LZ21H070001,LHDMD22H02001</funding_grant_id><funding_grant_id>SHSMU-ZDCX20212700</funding_grant_id><funding_grant_id>81722012</funding_grant_id><funding_grant_id>No.2025HZZD06</funding_grant_id><funding_grant_id>ZJ2022117</funding_grant_id><funding_grant_id>21SG11</funding_grant_id><funding_grant_id>2022ZZ01002</funding_grant_id><funding_grant_id>2018YFA0800403,2021YFC2701903</funding_grant_id><funding_grant_id>2022M723284</funding_grant_id><funding_grant_id>82425012,92457301,32471170,91857110,81722012,82325010,82300893,82500965</funding_grant_id><pubmed_authors>Shi M</pubmed_authors><pubmed_authors>Zhang R</pubmed_authors><pubmed_authors>Ding W</pubmed_authors><pubmed_authors>Qiu X</pubmed_authors><pubmed_authors>Wang Q</pubmed_authors><pubmed_authors>Piao HL</pubmed_authors><pubmed_authors>Zheng M</pubmed_authors><pubmed_authors>Guo G</pubmed_authors><pubmed_authors>Chen W</pubmed_authors><pubmed_authors>Wang W</pubmed_authors><pubmed_authors>Meng ZX</pubmed_authors><pubmed_authors>Wang RR</pubmed_authors><pubmed_authors>Jin L</pubmed_authors><pubmed_authors>Pan X</pubmed_authors><pubmed_authors>Yan J</pubmed_authors><pubmed_authors>Li C</pubmed_authors><pubmed_authors>Yan S</pubmed_authors><pubmed_authors>Chen H</pubmed_authors><pubmed_authors>Liang B</pubmed_authors><pubmed_authors>Zhang W</pubmed_authors><pubmed_authors>Hu C</pubmed_authors><pubmed_authors>Luo P</pubmed_authors><pubmed_authors>Yang Y</pubmed_authors><pubmed_authors>Ji X</pubmed_authors><pubmed_authors>Zhang Z</pubmed_authors><pubmed_authors>Fu H</pubmed_authors><pubmed_authors>Zhu L</pubmed_authors><pubmed_authors>Yan D</pubmed_authors><pubmed_authors>Wu QQ</pubmed_authors><pubmed_authors>Zhu S</pubmed_authors></additional><is_claimable>false</is_claimable><name>BAF60a-dependent chromatin remodeling preserves β cell function and contributes to the therapeutic benefits of GLP-1R agonists.</name><description>Impaired glucose-stimulated insulin secretion (GSIS) is a hallmark of β cell dysfunction in diabetes.
