Here, we show that RETREG3/FAM134C functions as a selective autophagy receptor for the type I BMP receptor (BMPRIA/ALK3) and recruits BMPRIA into LC3-containing autophagosomes for subsequent degradation. FAM134C-induced degradation diminishes the availability of BMP receptors and thus the strength of BMP signaling. Inhibition of autophagy through chemical means or knockdown of key autophagy regulators, ATG5 or Beclin-1, prevents BMPR1A degradation. Additionally, disruption of the putative LC3-interacting region (LIR) motif in FAM134C completely abolishes its interaction with LC3, ther</pubmed_abstract><journal>The EMBO journal</journal><pubmed_title>Reticulophagy receptor FAM134C restrains BMP receptor signaling.</pubmed_title><pmcid>PMC12669696</pmcid><funding_grant_id>31771546</funding_grant_id><funding_grant_id>31730057</funding_grant_id><funding_grant_id>LD21C070001</funding_grant_id><funding_grant_id>2022YFC3401500</funding_grant_id><funding_grant_id>LZ22C070001</funding_grant_id><funding_grant_id>TD2023017</funding_grant_id><funding_grant_id>32321002</funding_grant_id><funding_grant_id>U21A20356</funding_grant_id><pubmed_authors>Wu J</pubmed_authors><pubmed_authors>Gu S</pubmed_authors><pubmed_authors>Xu P</pubmed_authors><pubmed_authors>Zhao B</pubmed_authors><pubmed_authors>Cao J</pubmed_authors><pubmed_authors>Zhang H</pubmed_authors><pubmed_authors>Lin X</pubmed_authors><pubmed_authors>Liu T</pubmed_authors><pubmed_authors>Liu X</pubmed_authors><pubmed_authors>Feng XH</pubmed_authors><pubmed_authors>He Z</pubmed_authors><pubmed_authors>Sun Q</pubmed_authors><pubmed_authors>Zheng M</pubmed_authors><pubmed_authors>Han J</pubmed_authors><pubmed_authors>Jin J</pubmed_authors><pubmed_authors>Yu Y</pubmed_authors></additional><is_claimable>false</is_claimable><name>Reticulophagy receptor FAM134C restrains BMP receptor signaling.</name><description>FAM134/RETREG family members are ER-phagy receptors that maintain cellular homeostasis by regulating endoplasmic reticulum turnover. However, possible non-ER-phagy functions of FAM134 proteins remain elusive.
