It has been recently demonstrated in members of a Finnish family to co-segregate with heterozygosity for a frameshift variant in the fifth and last exon of the repressor element 1-silencing transcription factor gene (REST). Here, we report the first Italian family in which JS was diagnosed in the proband, a 38-year-old woman, and in her mother. Exome Sequencing identified in both, but not in clinically unaffected members of the family (i.e., a sister and the brother of the proband), the heterozygous pathogenic variant c.2645T>G (p.Leu882*) in exon-5 of the REST gene. This study confirms that exon-5 REST variants cause JS.</pubmed_abstract><journal>Clinical genetics</journal><pagination>199-203</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC12674978</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Novel Pathogenic Variant Confirms the Association of REST and Jones Syndrome.</pubmed_title><pmcid>PMC12674978</pmcid><pubmed_authors>Grammatico P</pubmed_authors><pubmed_authors>Turchetta R</pubmed_authors><pubmed_authors>Palmisano B</pubmed_authors><pubmed_authors>Riminucci M</pubmed_authors><pubmed_authors>Celli L</pubmed_authors><pubmed_authors>Allegra E</pubmed_authors><pubmed_authors>Lodato V</pubmed_authors><pubmed_authors>Colizza A</pubmed_authors><pubmed_authors>Bianco MR</pubmed_authors><pubmed_authors>Stanganelli FRF</pubmed_authors><pubmed_authors>Galli M</pubmed_authors><pubmed_authors>Corsi A</pubmed_authors><pubmed_authors>Bottillo I</pubmed_authors><pubmed_authors>Messineo D</pubmed_authors><pubmed_authors>D'Angeli G</pubmed_authors><pubmed_authors>Gianno F</pubmed_authors></additional><is_claimable>false</is_claimable><name>Novel Pathogenic Variant Confirms the Association of REST and Jones Syndrome.</name><description>Jones syndrome (JS) is an ultra-rare autosomal dominant condition characterized by gingival fibromatosis and progressive sensorineural hearing loss.
