While tetraspanin 4 (TSPAN4) is known to be expressed in the vasculature, its function in VSMC phenotypic switching and vascular disease is currently unknown. Here, we investigated the role of TSPAN4 using an in vitro model of platelet-derived growth factor BB (PDGF-BB)-induced phenotypic switching and an in vivo carotid artery ligation model in wildtype and TSPAN4-deficient mice. Our experiments, including EdU assays, Transwell assays, western blot analysis, and immunoprecipitation, revealed that TSPAN4 expression is elevated in human atherosclerotic arteries, ligated mouse carotid arteries, and PD</pubmed_abstract><journal>Clinical science (London, England : 1979)</journal><pubmed_title>TSPAN4 controls vascular smooth muscle cell phenotypic switching and intimal hyperplasia by targeting TPM1-regulated cytoskeletal organization.</pubmed_title><pmcid>PMC12687436</pmcid><funding_grant_id>2022YFS0627-A1</funding_grant_id><funding_grant_id>2021YJ0200</funding_grant_id><funding_grant_id>82030007, U23A20398</funding_grant_id><funding_grant_id>2022ZYD0057</funding_grant_id><funding_grant_id>2021ZKMS005</funding_grant_id><funding_grant_id>2022YFS0578, 2022YFS0614,2022YFS0607,2022YFS0627</funding_grant_id><funding_grant_id>00040155</funding_grant_id><funding_grant_id>2024NSFSC0580</funding_grant_id><pubmed_authors>Qiao G</pubmed_authors><pubmed_authors>Jiang J</pubmed_authors><pubmed_authors>Fu S</pubmed_authors><pubmed_authors>Li J</pubmed_authors><pubmed_authors>Chen K</pubmed_authors><pubmed_authors>Chen J</pubmed_authors><pubmed_authors>Yuan Q</pubmed_authors><pubmed_authors>Li R</pubmed_authors><pubmed_authors>Li M</pubmed_authors><pubmed_authors>Yi J</pubmed_authors><pubmed_authors>Zhang T</pubmed_authors><pubmed_authors>Li S</pubmed_authors><pubmed_authors>Feng J</pubmed_authors><pubmed_authors>Zhang Y</pubmed_authors><pubmed_authors>Jiang D</pubmed_authors><pubmed_authors>Zhang C</pubmed_authors><pubmed_authors>Yu Y</pubmed_authors></additional><is_claimable>false</is_claimable><name>TSPAN4 controls vascular smooth muscle cell phenotypic switching and intimal hyperplasia by targeting TPM1-regulated cytoskeletal organization.</name><description>Vascular smooth muscle cell (VSMC) phenotypic switching, followed by enhanced proliferation and migration, is a key event in the development of intimal hyperplasia in diverse vascular diseases.
