Ca2+ binding leads to conformation changes in CaM that enable engagement of a repertoire of enzymes and the regulation of their catalytic activities. Classically, Ca2+-CaM binds to an inhibitory pseudosubstrate sequence C-terminal to the kinase domain in members of the Ca2+-CaM-dependent protein kinase (CAMK) family and relieves inhibition to promote catalytic activity. Here, we report an unexpected mechanism by which CaM can bind CHK2 kinase to inhibit its kinase activity. Using biochemical, biophysical and structural mass spectrometry, we identify a direct interaction of Ca2+-CaM with the CHK2 kinase domain that suppresses CHK2 catalytic activity in vitro and identify K373 in CHK2 as crucial for cell prolifer</pubmed_abstract><journal>The Biochemical journal</journal><pubmed_title>Unconventional binding of calmodulin to CHK2 kinase inhibits catalytic activity.</pubmed_title><pmcid>PMC12751073</pmcid><funding_grant_id>1172929, 2034104, 9000719, 2034044, 2001817</funding_grant_id><pubmed_authors>Horne CR</pubmed_authors><pubmed_authors>Scott JW</pubmed_authors><pubmed_authors>Young SN</pubmed_authors><pubmed_authors>Burke JE</pubmed_authors><pubmed_authors>Petersen J</pubmed_authors><pubmed_authors>Belousoff MJ</pubmed_authors><pubmed_authors>Vaibhav V</pubmed_authors><pubmed_authors>Manning G</pubmed_authors><pubmed_authors>Dite TA</pubmed_authors><pubmed_authors>Suresh S</pubmed_authors><pubmed_authors>Wang T</pubmed_authors><pubmed_authors>Davies KA</pubmed_authors><pubmed_authors>Dagley LF</pubmed_authors><pubmed_authors>Nyvall HG</pubmed_authors><pubmed_authors>Murphy JM</pubmed_authors><pubmed_authors>Means AR</pubmed_authors><pubmed_authors>Gonzalez Castro A</pubmed_authors><pubmed_authors>Mather LJ</pubmed_authors></additional><is_claimable>false</is_claimable><name>Unconventional binding of calmodulin to CHK2 kinase inhibits catalytic activity.</name><description>Calmodulin (CaM) serves an essential role in eukaryotic cells as a Ca2+ sensor.
