IORPS was constructed based on the cumulative weighting of differentially expressed gene (DEG) expression levels. The predictive performance of conventional biomarkers, individual DEGs, and IORPS was evaluated for immunotherapy response and prognostic outcomes. The clinical relevance of IORPS was further validated using two external cohorts from the GEO database (CLB-IHN</pubmed_abstract><journal>Technology in cancer research &amp; treatment</journal><pagination>15330338251411026</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC12833181</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Immunotherapy Response Predictive Score Based on Tumor Microenvironment Profiles for Predicting Immunotherapy Outcomes in Advanced Head and Neck Cancer.</pubmed_title><pmcid>PMC12833181</pmcid><pubmed_authors>Chou MC</pubmed_authors><pubmed_authors>Chen LT</pubmed_authors><pubmed_authors>Pan MR</pubmed_authors><pubmed_authors>Cho SF</pubmed_authors><pubmed_authors>Wu CW</pubmed_authors><pubmed_authors>Tsai HT</pubmed_authors><pubmed_authors>Du JS</pubmed_authors><pubmed_authors>Hung YH</pubmed_authors><pubmed_authors>Chan LP</pubmed_authors><pubmed_authors>Wang HC</pubmed_authors><pubmed_authors>Wu CC</pubmed_authors><pubmed_authors>Liu YC</pubmed_authors><pubmed_authors>Moi SH</pubmed_authors></additional><is_claimable>false</is_claimable><name>Immunotherapy Response Predictive Score Based on Tumor Microenvironment Profiles for Predicting Immunotherapy Outcomes in Advanced Head and Neck Cancer.</name><description>ObjectivesThis retrospective study presents an integrative transcriptomic approach for recurrent and/or metastatic head and neck squamous cell carcinoma (R/M HNSCC) by developing an immune response predictive score (IORPS) derived from tumor microenvironment (TME) transcriptomic profiles.MethodsA total of 30 R/M HNSCC patients treated with pembrolizumab or nivolumab, with available immune TME profiling data, were analyzed.
