Research using various experimental systems has shown that Mfn-2 is a mediator of mitochondrial fusion, an evolutionarily conserved process responsible for the surveillance of mitochondrial homeostasis. Here, we find that cardiac myocyte mitochondria lacking Mfn-2 are pleiomorphic and have the propensity to become enlarged. Consistent with an underlying mild mitochondrial dysfunction, Mfn-2-deficient mice display modest cardiac hypertrophy accompanied by slight functional deterioration. The absence of Mfn-2 is associated with a marked delay in mitochondrial permeability transition downstream of Ca(2+) stimulation or due to local generation of reactive oxygen species (ROS</pubmed_abstract><journal>Molecular and cellular biology</journal><pubmed_title>Mitofusin-2 maintains mitochondrial structure and contributes to stress-induced permeability transition in cardiac myocytes.</pubmed_title><pmcid>PMC3067905</pmcid><funding_grant_id>HL064750</funding_grant_id><funding_grant_id>HL102874</funding_grant_id><funding_grant_id>R21 HL102874</funding_grant_id><funding_grant_id>N01HV28178</funding_grant_id><funding_grant_id>AG34972</funding_grant_id><funding_grant_id>R01 HL061639</funding_grant_id><funding_grant_id>P01 HL074237</funding_grant_id><funding_grant_id>AG15052</funding_grant_id><funding_grant_id>R01 AG034972</funding_grant_id><funding_grant_id>R37 AG015052</funding_grant_id><funding_grant_id>P01 HL068758</funding_grant_id><funding_grant_id>HL68758</funding_grant_id><funding_grant_id>R01 AG015052</funding_grant_id><funding_grant_id>HL061639</funding_grant_id><funding_grant_id>R01 HL064750</funding_grant_id><funding_grant_id>HL074237</funding_grant_id><pubmed_authors>Luptak I</pubmed_authors><pubmed_authors>Riley DD</pubmed_authors><pubmed_authors>Colucci WS</pubmed_authors><pubmed_authors>Stanley WC</pubmed_authors><pubmed_authors>Walsh K</pubmed_authors><pubmed_authors>Lederer WJ</pubmed_authors><pubmed_authors>Papanicolaou KN</pubmed_authors><pubmed_authors>O'Shea KM</pubmed_authors><pubmed_authors>Ngoh GA</pubmed_authors><pubmed_authors>Khairallah RJ</pubmed_authors><pubmed_authors>Chikando A</pubmed_authors><pubmed_authors>Lugus JJ</pubmed_authors></additional><is_claimable>false</is_claimable><name>Mitofusin-2 maintains mitochondrial structure and contributes to stress-induced permeability transition in cardiac myocytes.</name><description>Mitofusin-2 (Mfn-2) is a dynamin-like protein that is involved in the rearrangement of the outer mitochondrial membrane.
