Only 5-10% of the total observed variance is explained by the genetic loci found to date, suggesting that additional loci should be searched using approaches alternative to large meta analysis. GWAS (Genome Wide Association Study) for red blood cell traits in a founder population cohort from Northern Italy identified a new locus for mean corpuscular hemoglobin concentration (MCHC) in the TAF3 gene. The association was replicated in two cohorts (rs1887582, P = 4.25E-09). TAF3 encodes a transcription cofactor that participates in core promoter recognition complex, and is involved in zebrafish and mouse erythropoiesis. We show here that TAF3 is required for transcription of the SPTA1 gene, encoding a</pubmed_abstract><journal>PloS one</journal><pubmed_title>Genome wide association analysis of a founder population identified TAF3 as a gene for MCHC in humans.</pubmed_title><pmcid>PMC3729833</pmcid><funding_grant_id>R01GM033977</funding_grant_id><funding_grant_id>N01.AG. 12100</funding_grant_id><funding_grant_id>N01.AG.916413</funding_grant_id><funding_grant_id>R01 MD009164</funding_grant_id><funding_grant_id>N01.AG. 821336</funding_grant_id><funding_grant_id>T34 GM008574</funding_grant_id><funding_grant_id>263 MD 821336</funding_grant_id><funding_grant_id>G0000111</funding_grant_id><funding_grant_id>R01 GM033977</funding_grant_id><funding_grant_id>R01 HL105756</funding_grant_id><funding_grant_id>263 MD 9164 13</funding_grant_id><pubmed_authors>Toniolo D</pubmed_authors><pubmed_authors>Psaty BM</pubmed_authors><pubmed_authors>Rotter JI</pubmed_authors><pubmed_authors>Uitterlinden AG</pubmed_authors><pubmed_authors>Rice K</pubmed_authors><pubmed_authors>Boerwinkle E</pubmed_authors><pubmed_authors>Gudnason V</pubmed_authors><pubmed_authors>Masciullo C</pubmed_authors><pubmed_authors>Pirastu N</pubmed_authors><pubmed_authors>Gasparini P</pubmed_authors><pubmed_authors>Okonkwo SU</pubmed_authors><pubmed_authors>Pistis G</pubmed_authors><pubmed_authors>Spector TD</pubmed_authors><pubmed_authors>Hart D</pubmed_authors><pubmed_authors>Ferrucci L</pubmed_authors><pubmed_authors>Green MR</pubmed_authors><pubmed_authors>Oostra BA</pubmed_authors><pubmed_authors>Furth S</pubmed_authors><pubmed_authors>Hernandez D</pubmed_authors><pubmed_authors>Dehghan A</pubmed_authors><pubmed_authors>Kottgen A</pubmed_authors><pubmed_authors>Zakai NA</pubmed_authors><pubmed_authors>Patel KV</pubmed_authors><pubmed_authors>Massacane R</pubmed_authors><pubmed_authors>van Duijn CM</pubmed_authors><pubmed_authors>Traglia M</pubmed_authors><pubmed_authors>Verwoert G</pubmed_authors><pubmed_authors>Levy D</pubmed_authors><pubmed_authors>Harris TB</pubmed_authors><pubmed_authors>Cupples L</pubmed_authors><pubmed_authors>Yang Q</pubmed_authors><pubmed_authors>Lin J</pubmed_authors><pubmed_authors>Thein SL</pubmed_authors><pubmed_authors>Nalls M</pubmed_authors><pubmed_authors>Lumley T</pubmed_authors><pubmed_authors>Fox CS</pubmed_authors><pubmed_authors>Ganesh S</pubmed_authors><pubmed_authors>Eiriksdottir G</pubmed_authors><pubmed_authors>Aspelund T</pubmed_authors><pubmed_authors>Tanaka T</pubmed_authors><pubmed_authors>Hofman A</pubmed_authors><pubmed_authors>Mangino M</pubmed_authors><pubmed_authors>Ehret G</pubmed_authors><pubmed_authors>Smith AV</pubmed_authors><pubmed_authors>Zheng G</pubmed_authors><pubmed_authors>Soranzo N</pubmed_authors><pubmed_authors>Chakravarti A</pubmed_authors><pubmed_authors>Camaschella C</pubmed_authors><pubmed_authors>Singleton AB</pubmed_authors><pubmed_authors>Cushman M</pubmed_authors><pubmed_authors>Ganesh SK</pubmed_authors><pubmed_authors>Sala C</pubmed_authors><pubmed_authors>Jaffe A</pubmed_authors><pubmed_authors>CHARGE Consortium Hematology Working</pubmed_authors><pubmed_authors>Bis JC</pubmed_authors><pubmed_authors>Coresh J</pubmed_authors><pubmed_authors>van Rooij FJ</pubmed_authors><pubmed_authors>Witteman JC</pubmed_authors><pubmed_authors>Chen MH</pubmed_authors><pubmed_authors>Launer LJ</pubmed_authors><pubmed_authors>Buetti I</pubmed_authors><pubmed_authors>Glazer NL</pubmed_authors><pubmed_authors>Guralnik JM</pubmed_authors><pubmed_authors>O' Donnell CJ</pubmed_authors><pubmed_authors>Shin SY</pubmed_authors></additional><is_claimable>false</is_claimable><name>Genome wide association analysis of a founder population identified TAF3 as a gene for MCHC in humans.</name><description>The red blood cell related traits are highly heritable but their genetics are poorly defined.
