The CACNA1B variant c.4166G>A; (p.R1389H) was recently reported as the likely causative mutation in a single 3-generation Dutch pedigree with five subjects affected by a unique dominant M-D syndrome and cardiac arrhythmias. In an attempt to replicate this finding, we assessed by direct sequencing the frequency of CACNA1B c.4166G>A; (p.R1389H) in a cohort of 520 M-D cases, in which SGCE mutations had been previously excluded. A total of 146 cases (28%) had a positive family history of M-D. The frequency of the variant was also assessed in 489 neurologically healthy controls and in publicly available data sets of genetic variation (1000 Genomes, Exome Variant Server and Exome Aggregati</pubmed_abstract><journal>Human molecular genetics</journal><pubmed_title>The CACNA1B R1389H variant is not associated with myoclonus-dystonia in a large European multicentric cohort.</pubmed_title><pmcid>PMC4550822</pmcid><funding_grant_id>RC2 HL102926</funding_grant_id><funding_grant_id>NF-SI-0507-10376</funding_grant_id><funding_grant_id>RC2 HL102925</funding_grant_id><funding_grant_id>UC2 HL102923</funding_grant_id><funding_grant_id>RC2 HL102924</funding_grant_id><funding_grant_id>RC2 HL102923</funding_grant_id><funding_grant_id>WT089698/Z/09/Z</funding_grant_id><funding_grant_id>UC2 HL102924</funding_grant_id><funding_grant_id>MC_G1000735</funding_grant_id><funding_grant_id>UC2 HL102925</funding_grant_id><funding_grant_id>UC2 HL102926</funding_grant_id><funding_grant_id>RC2 HL103010</funding_grant_id><funding_grant_id>NF-SI-0513-10064</funding_grant_id><funding_grant_id>089698</funding_grant_id><funding_grant_id>UC2 HL103010</funding_grant_id><pubmed_authors>Nardocci N</pubmed_authors><pubmed_authors>Pittman AM</pubmed_authors><pubmed_authors>Hardy J</pubmed_authors><pubmed_authors>Carecchio M</pubmed_authors><pubmed_authors>Zorzi G</pubmed_authors><pubmed_authors>Gasser T</pubmed_authors><pubmed_authors>Bandres-Ciga S</pubmed_authors><pubmed_authors>Mencacci NE</pubmed_authors><pubmed_authors>Wood NW</pubmed_authors><pubmed_authors>Garavaglia B</pubmed_authors><pubmed_authors>R'bibo L</pubmed_authors><pubmed_authors>Klein C</pubmed_authors><pubmed_authors>Bhatia KP</pubmed_authors><pubmed_authors>Weissbach A</pubmed_authors><pubmed_authors>Lohmann E</pubmed_authors><pubmed_authors>Batla A</pubmed_authors></additional><is_claimable>false</is_claimable><name>The CACNA1B R1389H variant is not associated with myoclonus-dystonia in a large European multicentric cohort.</name><description>Myoclonus-dystonia (M-D) is a very rare movement disorder, caused in ∼30-50% of cases by mutations in SGCE.
