The galactolipid synthases MGD1 and DGD1 catalyze consecutive galactosyltransfer reactions but localize to the inner and outer chloroplast envelopes, respectively, necessitating intermembrane lipid transfer. Here we show that the N-terminal sequence of DGD1 (NDGD1) is required for galactolipid transfer between the envelopes. Different diglycosyllipid synthases (DGD1, DGD2, and Chloroflexus glucosyltransferase) were introduced into the dgd1-1 mutant of Arabidopsis in fusion with N-terminal extensions (NDGD1 and NDGD2) targeting to the outer envelope. Reconstruction of DGDG synthesis in the outer envelope membrane was observed only with diglycosyllipi</pubmed_abstract><journal>Proceedings of the National Academy of Sciences of the United States of America</journal><pubmed_title>Synthesis and transfer of galactolipids in the chloroplast envelope membranes of Arabidopsis thaliana.</pubmed_title><pmcid>PMC5035899</pmcid><funding_grant_id>DE-FG02-98ER20305</funding_grant_id><funding_grant_id>Fellowship</funding_grant_id><funding_grant_id>SFB645</funding_grant_id><funding_grant_id>Do520/10</funding_grant_id><pubmed_authors>Melzer M</pubmed_authors><pubmed_authors>Kelly AA</pubmed_authors><pubmed_authors>Schulze S</pubmed_authors><pubmed_authors>Roston RL</pubmed_authors><pubmed_authors>Kalisch B</pubmed_authors><pubmed_authors>Thiele J</pubmed_authors><pubmed_authors>Dormann P</pubmed_authors><pubmed_authors>Holzl G</pubmed_authors><pubmed_authors>Benning C</pubmed_authors></additional><is_claimable>false</is_claimable><name>Synthesis and transfer of galactolipids in the chloroplast envelope membranes of Arabidopsis thaliana.</name><description>Galactolipids [monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG)] are the hallmark lipids of photosynthetic membranes.
