We have previously reported that overexpressed SPL displays anti-influenza viral activity; however, the underlying mechanism is incompletely understood. In this study, we demonstrate that SPL functions as a positive regulator of IKKε to propel type I IFN-mediated innate immune responses against viral infection. Exogenous SPL expression inhibited influenza A virus replication, which correlated with an increase in type I IFN production and IFN-stimulated gene accumulation upon infection. In contrast, the lack of SPL expression led to an elevated cellular susceptibility to influenza A virus infection. In support of this, SPL-deficient cells were defective</pubmed_abstract><journal>Journal of immunology (Baltimore, Md. : 1950)</journal><pubmed_title>Sphingosine 1-Phosphate Lyase Enhances the Activation of IKKε To Promote Type I IFN-Mediated Innate Immune Responses to Influenza A Virus Infection.</pubmed_title><pmcid>PMC5528153</pmcid><funding_grant_id>R56 AI108861</funding_grant_id><funding_grant_id>R01 GM110776</funding_grant_id><funding_grant_id>R01 AI091797</funding_grant_id><pubmed_authors>Hiscott J</pubmed_authors><pubmed_authors>Vijayan M</pubmed_authors><pubmed_authors>Fox TE</pubmed_authors><pubmed_authors>Hahm B</pubmed_authors><pubmed_authors>Ngo H</pubmed_authors><pubmed_authors>Johnson MC</pubmed_authors><pubmed_authors>Alexander S</pubmed_authors><pubmed_authors>Kester M</pubmed_authors><pubmed_authors>Drews K</pubmed_authors><pubmed_authors>Xia C</pubmed_authors><pubmed_authors>Song YE</pubmed_authors><pubmed_authors>Studstill CJ</pubmed_authors></additional><is_claimable>false</is_claimable><name>Sphingosine 1-Phosphate Lyase Enhances the Activation of IKKε To Promote Type I IFN-Mediated Innate Immune Responses to Influenza A Virus Infection.</name><description>Sphingosine 1-phosphate (S1P) lyase (SPL) is an intracellular enzyme that mediates the irreversible degradation of the bioactive lipid S1P.
