We found that intracellular target engagement of unlabeled new therapeutics can be quantitated using polarized microscopy combined with competitive binding of matched fluorescent companion imaging probes. We quantitated the dynamics of target engagement of covalent BTK inhibitors, as well as reversible PARP inhibitors, in populations of single cells using a single companion imaging probe for each target. We then determined average in vivo tumor concentrations and found marked population heterogeneity following systemic delivery, revealing single cells with low target occupancy at high average target engagement in vivo.</pubmed_abstract><journal>Nature chemical biology</journal><pubmed_title>Quantitating drug-target engagement in single cells in vitro and in vivo.</pubmed_title><pmcid>PMC5630128</pmcid><funding_grant_id>R01 CA241179</funding_grant_id><funding_grant_id>T32 CA079443</funding_grant_id><funding_grant_id>R01 HL122208</funding_grant_id><funding_grant_id>P50 CA086355</funding_grant_id><funding_grant_id>R01 CA164448</funding_grant_id><funding_grant_id>K99 CA198857</funding_grant_id><funding_grant_id>R01 EB010011</funding_grant_id><funding_grant_id>R00 CA198857</funding_grant_id><pubmed_authors>Cuccarese M</pubmed_authors><pubmed_authors>Giedt RJ</pubmed_authors><pubmed_authors>Yang K</pubmed_authors><pubmed_authors>Kim E</pubmed_authors><pubmed_authors>Weissleder R</pubmed_authors><pubmed_authors>Vinegoni C</pubmed_authors><pubmed_authors>Dubach JM</pubmed_authors><pubmed_authors>Meimetis LG</pubmed_authors></additional><is_claimable>false</is_claimable><name>Quantitating drug-target engagement in single cells in vitro and in vivo.</name><description>Quantitation of drug target engagement in single cells has proven to be difficult, often leaving unanswered questions in the drug development process.
