Simple high-throughput technologies are desperately needed to delineate the landscape of PPI networks. While recent state-of-the-art yeast two-hybrid (Y2H) systems improved screening efficiency, either individual colony isolation, library preparation arrays, gene barcoding or massive sequencing are still required. Here, we developed a recombination-based 'library vs library' Y2H system (RLL-Y2H), by which multi-library screening can be accomplished in a single pool without any individual treatment. This system is based on the phiC31 integrase-mediated integration between bait and prey plasmids. The integrated fragments were digested by MmeI and subjected to deep sequencing to decode the interaction matrix</pubmed_abstract><journal>Nucleic acids research</journal><pagination>e17</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC5815087</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Development and application of a recombination-based library versus library high- throughput yeast two-hybrid (RLL-Y2H) screening system.</pubmed_title><pmcid>PMC5815087</pmcid><pubmed_authors>Zhou M</pubmed_authors><pubmed_authors>Habib Z</pubmed_authors><pubmed_authors>Cao G</pubmed_authors><pubmed_authors>Wang D</pubmed_authors><pubmed_authors>Fu Z</pubmed_authors><pubmed_authors>Zhong W</pubmed_authors><pubmed_authors>Han Y</pubmed_authors><pubmed_authors>Yao Q</pubmed_authors><pubmed_authors>Chen H</pubmed_authors><pubmed_authors>Xiao S</pubmed_authors><pubmed_authors>Yang F</pubmed_authors><pubmed_authors>Rahman K</pubmed_authors><pubmed_authors>Zhu C</pubmed_authors><pubmed_authors>Lin D</pubmed_authors><pubmed_authors>Lei Y</pubmed_authors><pubmed_authors>Tao R</pubmed_authors><pubmed_authors>Chen X</pubmed_authors><pubmed_authors>Xu W</pubmed_authors><pubmed_authors>Tyagi R</pubmed_authors><pubmed_authors>Wu X</pubmed_authors><pubmed_authors>Yu Y</pubmed_authors></additional><is_claimable>false</is_claimable><name>Development and application of a recombination-based library versus library high- throughput yeast two-hybrid (RLL-Y2H) screening system.</name><description>Protein-protein interaction (PPI) network maintains proper function of all organisms.
