We selected a severe early onset form of Leber congenital amaurosis (LCA), caused by mutations in the gene LCA5, in order to test the efficacy of gene augmentation therapy for a ciliopathy. The LCA5-encoded protein, Lebercilin, is essential for the trafficking of proteins and vesicles to the photoreceptor outer segment. Using the AAV serotype AAV7m8 to deliver a human LCA5 cDNA into an Lca5 null mouse model of LCA5, we show partial rescue of retinal structure and visual function. Specifically, we observed restoration of rod-and-cone-driven electroretinograms in about 25% of injected eyes, restoration of pupillary light responses in the majority of treated eyes, an ∼20-fold decrease in target </pubmed_abstract><journal>Molecular therapy : the journal of the American Society of Gene Therapy</journal><pubmed_title>Amelioration of Neurosensory Structure and Function in Animal and Cellular Models of a Congenital Blindness.</pubmed_title><pmcid>PMC5986734</pmcid><funding_grant_id>P30 EY001583</funding_grant_id><funding_grant_id>8DP1EY023177</funding_grant_id><funding_grant_id>R24 EY019861</funding_grant_id><funding_grant_id>1R24 EY019861-01A1</funding_grant_id><funding_grant_id>DP1 EY023177</funding_grant_id><pubmed_authors>Aleman TS</pubmed_authors><pubmed_authors>Cremers FPM</pubmed_authors><pubmed_authors>Pham J</pubmed_authors><pubmed_authors>den Hollander AI</pubmed_authors><pubmed_authors>McNamara I</pubmed_authors><pubmed_authors>Nishina P</pubmed_authors><pubmed_authors>Aravand P</pubmed_authors><pubmed_authors>Bennett DJ</pubmed_authors><pubmed_authors>Wei Z</pubmed_authors><pubmed_authors>Song JY</pubmed_authors><pubmed_authors>Lyubarsky A</pubmed_authors><pubmed_authors>Sun J</pubmed_authors><pubmed_authors>Pan W</pubmed_authors><pubmed_authors>de Melo J</pubmed_authors><pubmed_authors>Zhou S</pubmed_authors><pubmed_authors>Maguire AM</pubmed_authors><pubmed_authors>Roepman R</pubmed_authors><pubmed_authors>Herrera P</pubmed_authors><pubmed_authors>Bennett J</pubmed_authors><pubmed_authors>Mills J</pubmed_authors><pubmed_authors>Pan J</pubmed_authors><pubmed_authors>Shpylchak I</pubmed_authors><pubmed_authors>Nikonov S</pubmed_authors><pubmed_authors>Ying GS</pubmed_authors><pubmed_authors>Bennicelli JL</pubmed_authors><pubmed_authors>Leo L</pubmed_authors><pubmed_authors>Koenekoop RK</pubmed_authors><pubmed_authors>Commins N</pubmed_authors></additional><is_claimable>false</is_claimable><name>Amelioration of Neurosensory Structure and Function in Animal and Cellular Models of a Congenital Blindness.</name><description>Most genetically distinct inherited retinal degenerations are primary photoreceptor degenerations.
