Renal dysfunction is associated with crizotinib therapy but the mechanism is unknown. Here, we report a case of anaplastic lymphoma kinase positive non-small cell lung cancer showing multiple cysts and dysfunction of the kidneys during crizotinib administration. We also present results demonstrating that long-term crizotinib treatment induces fibrosis and dysfunction of the kidneys by activating the tumor necrosis factor-α/nuclear factor-κB signaling pathway. In conclusion, this study shows the renal detrimental effects of crizotinib, suggesting the need of careful monitoring of renal function during crizotinib therapy.</pubmed_abstract><journal>International journal of molecular sciences</journal><pagination>E2902</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC6213486</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Renal Injury during Long-Term Crizotinib Therapy.</pubmed_title><pmcid>PMC6213486</pmcid><pubmed_authors>Baffour Tonto P</pubmed_authors><pubmed_authors>Yonemura S</pubmed_authors><pubmed_authors>Kobayashi T</pubmed_authors><pubmed_authors>Nishihama K</pubmed_authors><pubmed_authors>Toda M</pubmed_authors><pubmed_authors>Takeshita A</pubmed_authors><pubmed_authors>Fujimoto H</pubmed_authors><pubmed_authors>Taguchi O</pubmed_authors><pubmed_authors>Yasuma T</pubmed_authors><pubmed_authors>Ito K</pubmed_authors><pubmed_authors>D'Alessandro-Gabazza CN</pubmed_authors><pubmed_authors>Gabazza EC</pubmed_authors><pubmed_authors>Nishii Y</pubmed_authors><pubmed_authors>Hataji O</pubmed_authors></additional><is_claimable>false</is_claimable><name>Renal Injury during Long-Term Crizotinib Therapy.</name><description>Crizotinib is highly effective against anaplastic lymphoma kinase-positive and c-ros oncogen1-positive non-small cell lung cancer.
