The retinal pigment epithelium (RPE) is a primary site of disease pathogenesis. The genetic basis of AMD is relatively well understood; however, this knowledge is yet to yield a treatment for the most prevalent non-neovascular disease forms. Therefore, tissue-specific epigenetic mechanisms of gene regulation are of considerable interest in AMD. We aimed to identify differentially methylated genes associated with AMD in the RPE and differentiate local DNA methylation aberrations from global DNA methylation changes, as local DNA methylation changes may be more amenable to therapeutic manipulation.<h4>Methods</h4>Epigenome-wide association study and t</pubmed_abstract><journal>Clinical epigenetics</journal><pubmed_title>Whole-genome methylation profiling of the retinal pigment epithelium of individuals with age-related macular degeneration reveals differential methylation of the SKI, GTF2H4, and TNXB genes.</pubmed_title><pmcid>PMC6332695</pmcid><funding_grant_id>SGL019\1076</funding_grant_id><funding_grant_id>ref 1732/1733</funding_grant_id><funding_grant_id>NIHR</funding_grant_id><funding_grant_id>MR/K024418/1</funding_grant_id><funding_grant_id>R21EY028273-01A1</funding_grant_id><funding_grant_id>CL-2015-07-004</funding_grant_id><funding_grant_id>R21 EY028273</funding_grant_id><funding_grant_id>1732/33</funding_grant_id><funding_grant_id>24NE141</funding_grant_id><funding_grant_id>SAC025</funding_grant_id><pubmed_authors>Fang Y</pubmed_authors><pubmed_authors>Rathi S</pubmed_authors><pubmed_authors>Saptarshi N</pubmed_authors><pubmed_authors>de Jong EK</pubmed_authors><pubmed_authors>Paraoan L</pubmed_authors><pubmed_authors>den Hollander AI</pubmed_authors><pubmed_authors>Bishop PN</pubmed_authors><pubmed_authors>Liloglou T</pubmed_authors><pubmed_authors>Olsen TW</pubmed_authors><pubmed_authors>Clark SJ</pubmed_authors><pubmed_authors>Porter LF</pubmed_authors><pubmed_authors>Chavali VRM</pubmed_authors></additional><is_claimable>false</is_claimable>

<h4>Background</h4>Age-related macular degeneration (AMD) is a degenerative disorder of the central retina and the foremost cause of blindness.
