The molecular machinery and biological significance of M cell differentiation, however, remain to be fully elucidated. Here, we demonstrate that Sox8, a member of the SRY-related HMG box transcription factor family, is specifically expressed by M cells in the intestinal epithelium. The expression of Sox8 requires activation of RANKL-RelB signaling. Chromatin immunoprecipitation and luciferase assays revealed that Sox8 directly binds the promoter region of Gp2 to increase Gp2 expression, which is the hallmark of functionally mature M cells. Furthermore, genetic deletion of Sox8 causes a marked decrease in the </pubmed_abstract><journal>The Journal of experimental medicine</journal><pubmed_title>Sox8 is essential for M cell maturation to accelerate IgA response at the early stage after weaning in mice.</pubmed_title><pmcid>PMC6446867</pmcid><funding_grant_id>JP16H06598</funding_grant_id><funding_grant_id>JP16J09413</funding_grant_id><funding_grant_id>JP25460261</funding_grant_id><funding_grant_id>JP17H04089</funding_grant_id><funding_grant_id>JP25293114</funding_grant_id><funding_grant_id>JP16K08457</funding_grant_id><pubmed_authors>Obata Y</pubmed_authors><pubmed_authors>Takahashi D</pubmed_authors><pubmed_authors>Kimura S</pubmed_authors><pubmed_authors>Kobayashi N</pubmed_authors><pubmed_authors>Mutoh M</pubmed_authors><pubmed_authors>Sato S</pubmed_authors><pubmed_authors>Nakamura Y</pubmed_authors><pubmed_authors>Kato N</pubmed_authors><pubmed_authors>Fujiki R</pubmed_authors><pubmed_authors>Nakagawa T</pubmed_authors><pubmed_authors>Kanaya T</pubmed_authors><pubmed_authors>Iwanaga T</pubmed_authors><pubmed_authors>Kaisho T</pubmed_authors><pubmed_authors>Ohno H</pubmed_authors><pubmed_authors>Hase K</pubmed_authors></additional><is_claimable>false</is_claimable><name>Sox8 is essential for M cell maturation to accelerate IgA response at the early stage after weaning in mice.</name><description>Microfold (M) cells residing in the follicle-associated epithelium (FAE) of the gut-associated lymphoid tissue are specialized for antigen uptake to initiate mucosal immune responses.
