However, the impact of pretreatment variables, such as CD19 expression level, on leukemic blasts, the presence of CD19- subpopulations, and especially prior CD19-targeted therapy, on the response to CAR T-cell therapy has not been determined. We analyzed 166 patients treated with CAR T-cell therapy at our institution. Eleven patients did not achieve a minimal residual disease (MRD)- deep remission, whereas 67 patients had a recurrence after achieving a MRD- deep remission: 28 patients with CD19+ leukemia and 39 patients with CD19- leukemia. Return of CD19+ leukemia was associated with loss of CAR T-cell function, whereas CD19- le</pubmed_abstract><journal>Blood advances</journal><pubmed_title>CAR T-cell therapy is effective for CD19-dim B-lymphoblastic leukemia but is impacted by prior blinatumomab therapy.</pubmed_title><pmcid>PMC6880911</pmcid><funding_grant_id>P30 CA016520</funding_grant_id><funding_grant_id>P01 CA214278</funding_grant_id><funding_grant_id>U01 CA232563</funding_grant_id><pubmed_authors>Pillai V</pubmed_authors><pubmed_authors>Muralidharan K</pubmed_authors><pubmed_authors>Cherian S</pubmed_authors><pubmed_authors>Oldridge DA</pubmed_authors><pubmed_authors>Mohan D</pubmed_authors><pubmed_authors>Rheingold SR</pubmed_authors><pubmed_authors>Melenhorst JJ</pubmed_authors><pubmed_authors>Meng W</pubmed_authors><pubmed_authors>Bhoj VG</pubmed_authors><pubmed_authors>DiNofia AM</pubmed_authors><pubmed_authors>Fromm JR</pubmed_authors><pubmed_authors>Paessler M</pubmed_authors><pubmed_authors>June CH</pubmed_authors><pubmed_authors>Thomas-Tikhonenko A</pubmed_authors><pubmed_authors>Luo M</pubmed_authors><pubmed_authors>Van Arnam J</pubmed_authors><pubmed_authors>Bagashev A</pubmed_authors><pubmed_authors>Grupp SA</pubmed_authors><pubmed_authors>Maude SL</pubmed_authors><pubmed_authors>Rosenthal J</pubmed_authors><pubmed_authors>Luning Prak ET</pubmed_authors><pubmed_authors>Wertheim G</pubmed_authors></additional><is_claimable>false</is_claimable><name>CAR T-cell therapy is effective for CD19-dim B-lymphoblastic leukemia but is impacted by prior blinatumomab therapy.</name><description>Tisagenlecleucel, a chimeric antigen receptor (CAR) T-cell product targeting CD19 is approved for relapsed/refractory B-cell acute lymphoblastic leukemia (B-ALL).
