The Ig-like domain-containing protein myotilin provides structural integrity to Z-discs-the boundaries between adjacent sarcomeres. Myotilin binds to Z-disc components, including F-actin and α-actinin-2, but the molecular mechanism of binding and implications of these interactions on Z-disc integrity are still elusive. To illuminate them, we used a combination of small-angle X-ray scattering, cross-linking mass spectrometry, and biochemical and molecular biophysics approaches. We discovered that myotilin displays conformational ensembles in solution. We generated a structural model of the F-actin:myotilin complex that r</pubmed_abstract><journal>PLoS biology</journal><pubmed_title>Molecular basis of F-actin regulation and sarcomere assembly via myotilin.</pubmed_title><pmcid>PMC8062120</pmcid><funding_grant_id>238423</funding_grant_id><funding_grant_id>201543</funding_grant_id><funding_grant_id>FOR 1352, FOR 2743</funding_grant_id><funding_grant_id>Collaborative Award (201543/Z/16)</funding_grant_id><funding_grant_id>I525, I1593, P22276, P19060 and W1221</funding_grant_id><funding_grant_id>35337, P1-0140</funding_grant_id><funding_grant_id>I 525</funding_grant_id><funding_grant_id>253275</funding_grant_id><funding_grant_id>Chemical Biology project LS17-008</funding_grant_id><funding_grant_id>201543/Z/16</funding_grant_id><funding_grant_id>390939984</funding_grant_id><funding_grant_id>I 1593</funding_grant_id><funding_grant_id>Austrian-Slovak Interreg Project B301</funding_grant_id><pubmed_authors>Sajko S</pubmed_authors><pubmed_authors>van der Ven PFM</pubmed_authors><pubmed_authors>Kostan J</pubmed_authors><pubmed_authors>Djinovic-Carugo K</pubmed_authors><pubmed_authors>Pavsic M</pubmed_authors><pubmed_authors>Molt S</pubmed_authors><pubmed_authors>Drepper F</pubmed_authors><pubmed_authors>Svergun DI</pubmed_authors><pubmed_authors>Lenarcic B</pubmed_authors><pubmed_authors>Schwarz TC</pubmed_authors><pubmed_authors>Graewert MA</pubmed_authors><pubmed_authors>Puz V</pubmed_authors><pubmed_authors>Konrat R</pubmed_authors><pubmed_authors>Furst DO</pubmed_authors><pubmed_authors>Warscheid B</pubmed_authors><pubmed_authors>Onipe A</pubmed_authors><pubmed_authors>Schreiner C</pubmed_authors></additional><is_claimable>false</is_claimable><name>Molecular basis of F-actin regulation and sarcomere assembly via myotilin.</name><description>Sarcomeres, the basic contractile units of striated muscle cells, contain arrays of thin (actin) and thick (myosin) filaments that slide past each other during contraction.
