Furthermore, BBB impairment and pericyte loss in Alzheimer's disease (AD) is well documented. We found that PDGF-BB:PDGFRβ signalling components were altered in human AD brains, with a marked reduction in vascular PDGFB. We hypothesised that reduced PDGF-BB:PDGFRβ signalling in pericytes may impact on the BBB. We therefore tested the effects of PDGF-BB on primary human brain pericytes in vitro to define pathways related to BBB function. Using pharmacological inhibitors, we dissected distinct aspects of the PDGF-BB response that are controlled by extracellular signal-regulated kinase (ERK) and Akt </pubmed_abstract><journal>Communications biology</journal><pagination>235</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC8931009</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Characterisation of PDGF-BB:PDGFRβ signalling pathways in human brain pericytes: evidence of disruption in Alzheimer's disease.</pubmed_title><pmcid>PMC8931009</pmcid><pubmed_authors>Wu J</pubmed_authors><pubmed_authors>Correia J</pubmed_authors><pubmed_authors>Heppner P</pubmed_authors><pubmed_authors>Faull RLM</pubmed_authors><pubmed_authors>Handley R</pubmed_authors><pubmed_authors>Mee EW</pubmed_authors><pubmed_authors>Johnson R</pubmed_authors><pubmed_authors>Coppieters N</pubmed_authors><pubmed_authors>Schweder P</pubmed_authors><pubmed_authors>Turner C</pubmed_authors><pubmed_authors>Tan A</pubmed_authors><pubmed_authors>Curtis MA</pubmed_authors><pubmed_authors>Li S</pubmed_authors><pubmed_authors>Singh-Bains MK</pubmed_authors><pubmed_authors>Jansson D</pubmed_authors><pubmed_authors>Rustenhoven J</pubmed_authors><pubmed_authors>Dragunow M</pubmed_authors><pubmed_authors>Narayan P</pubmed_authors><pubmed_authors>Park TI</pubmed_authors><pubmed_authors>Highet B</pubmed_authors><pubmed_authors>Aalderink M</pubmed_authors><pubmed_authors>Smyth LCD</pubmed_authors></additional><is_claimable>false</is_claimable><name>Characterisation of PDGF-BB:PDGFRβ signalling pathways in human brain pericytes: evidence of disruption in Alzheimer's disease.</name><description>Platelet-derived growth factor-BB (PDGF-BB):PDGF receptor-β (PDGFRβ) signalling in brain pericytes is critical to the development, maintenance and function of a healthy blood-brain barrier (BBB).
